摘 要:目的观察腹腔内注射活性氧(ROS)清除剂苯亚甲基叔丁基氮氧化物(PBN)对背根神经节慢性压迫(CCD)模型大鼠手术侧足底疼痛的影响,以阐明ROS在CCD大鼠疼痛中的作用。方法将33只成年雌性Sprague-Dawley大鼠随机分为正常对照组(6只)、假手术组(6只)和CCD模型组(21只);术后第7天,再将CCD模型组大鼠随机分为CCD+PBN100亚组(9只)、CCD+PBN30亚组(6只)和CCD+0.9%氯化钠溶液亚组(CCD+N亚组,6只)。应用vonFrey纤维丝分别检测术前机械刺激缩足反应阈值(PWMT)基础值(0d)和术后第1、3、5、7天大鼠损伤侧后足的PWMT。术后第7天,分别在CCD+PBN100亚组、CCD+PBN30亚组和CCD+N亚组大鼠腹腔内注射PBN100mg/kg或PBN30mg/kg或等体积0.9%氯化钠溶液,并于注射后0.5、1、2、4、6、8、24h检测大鼠损伤侧后足的PWMT。结果假手术组大鼠术后第1天和CCD模型组大鼠术后第1、3、5、7天的PWMT均较同组术前基础值和正常对照组同时间点显著降低(P值均<0.05),CCD模型组大鼠术后第1、3、5、7天的PWMT均较假手术组同时间点显著降低(P值均<0.05)。CCD+PBN100亚组和CCD+PBN30亚组大鼠注射药物后0.5、1、2h的PWMT均较CCD+N亚组同时间点显著升高(P值均<0.05),CCD+PBN100亚组大鼠注射药物后0.5、1h的PWMT均较CCD+PBN30亚组同时间点显著升高(P值均<0.05)。结论腹腔内注射ROS清除剂PBN可显著缓解CCD模型大鼠痛觉过敏,高剂量PBN(100mg/kg)缓解疼痛的效果更佳,表明ROS可能参与CCD模型大鼠疼痛信息的调节。Objective To evaluate the effect of intraperitoneal injection of Phenyl N-tert-butylnitrone (PBN), a free radical scavenger of reactive oxygen species (ROS), on footplate pain caused by chronic compression of dorsal root ganglia (CCD) and to describe the role of ROS in CCD pain in rats. Methods Thirty- three adult female Sprague-Dawley rats were randomly divided into normal blank group (n = 6), sham operation group (n=6) and CCD group (n=21). PBN 100 mg/kg (groupCCD+PBN100, n=9), PBN30 mg/kg (group CCD+ PBN30, n = 6) and the same volume of normal saline (group CCD + N, n = 6) were intraperitoneally injected in CCD rats, respectively. The paw withdrawal mechanical threshold (PWMT) was measured by von Frey hair before surgery, day 1, 3, 5 and 7 after surgery, and 0.5, 1, 2, 4, 6, 8 and 24 h after drug administration. Results Compared with PWMT before surgery, PWMT in sham operation group on day 1 after surgery and in CCD group on day 1, 2, 5 and 7 after surgery were significantly decreased (all P〈0. 05). PWMT in CCD group was significantly lower than that in sham operation group on day 1, 3, 5 and 7 after surgery (all P〈0. 05). PWMT in sham operation group on day 1 after surgery and in CCD group on day 1, 3, 5 and 7 after surgery were significantly lower than those in control group (all P〈,0.05). PWMT in group COD + PlaN100 and, group CCD + PBN30 were significantly increased as compared with group COD+ N at 0.5, 1, and 2 h after drug administration (all P〈0.05). PWMT in group COD+ PBN100 was significantly higher than that in group COD+ PBN30 at 0.5 and 1 h after drug administration (both P〈0.05). Conclusion PBN, especially high dose one (100 mg/kg), can alleviate pain in COD rats, which indicates that ROS may participate in the regulation of neuralgia pain.
