摘  要:探讨了超低温保存过程中预培养的蔗糖含量、DMSO含量,冰冻和解冻方式、防冻剂的种类和含量等因子对三尖杉悬浮细胞保存效果的影响.结果表明:效果较好的防冻剂为DMSO、PEG;在含有4%蔗糖和6%DMSO的液体培养基上培养得到的悬浮细胞,加入15%PEG、9%DMSO和5%山梨醇的防冻剂,采用快冻和缓解处理的悬浮细胞活力最强.The effects of several factors in cryopreservation process of Cephalataxus fortunei suspension cells were investigated, which included sugar concentration in preculture, freezing way, thawing way, kinds and concentration of cryoprotectant. The results showed that the better cryoprotectant was DMSO and PEG solution. The following cryopreserved C. fortunei suspension cells pos- sessed strongest vitality, which were precultured in 4% sucrose and 6% DMSO liquid, with the cryoprotectant of 15% DMSO, 9% PEP, and 5% sorbitol, frozon sharply and thowed gently.
