摘 要:目的研究1,3-二苯-1,3-丙二酮(1,3-diphenyl-1,3-proanedione,DPPD)对环磷酰胺诱导小鼠骨髓细胞微核的保护作用和对小鼠骨髓细胞微核的影响。方法 40只小鼠随机分为8组,分别为阴性对照组,环磷酰胺阳性对照组,环磷酰胺+DPPD低、中、高剂量组(62.5,250,1000 mg/kg),DPPD低、中、高剂量组(62.5,250,1000mg/kg),每组各5只。DPPD各剂量组小鼠经口给药30 d,每天1次。环磷酰胺采用30 h经口灌胃染毒。第2次染毒6 h后,小鼠麻醉脱臼处死,检测小鼠骨髓细胞微核率(千分率)。结果与阴性对照组比较,环磷酰胺阳性组小鼠骨髓细胞微核率明显升高(P<0.01),DPPD剂量组小鼠骨髓细胞微核率变化不明显(P>0.05);与环磷酰胺阳性组比较,环磷酰胺+DPPD低剂量组(62.5 mg/kg)小鼠骨髓细胞微核率变化不明显(P>0.05),环磷酰胺+DPPD中、高剂量组(250、1000 mg/kg)小鼠骨髓细胞微核率明显降低(P<0.01),微核抑制率分别为35.58%和61.54%。结论 30 d经口给予DPPD对小鼠骨髓细胞微核率没有影响,DPPD对环磷酰胺诱导小鼠骨髓细胞微核具有一定的预防保护作用。Objective To study the protective effect of 1,3-diphenyl-1,3-proanedione(DPPD) on bone marrow mi cronucleus induced by cyclophosphamide and the effects of DPPD on bone marrow micronucleus in mice.Methods Forty mice were divided eight groups randomLy,which were control group,cyclophosphamide positive control group,DPPD(62.5,250,1000 mg/kg) combined with cyclophosphamide groups,DPPD(62.5,250,1000 mg/kg) groups with 5 rats in each group.DPPD was administered orally once a day to the experimental groups for thirty days and Cy clophosphamide was administered by oral perfusion.The anesthetized mice were killed after the second administration to 6 hours and then examined the rate of bone marrow micronucleus.Results Compared with the control group,only the micronucleus rate of cyclophosphamide group increased obviously(P 0.01),and the micronucleus rates of DPPD groups had no significant difference.Compared with the cyclophosphamide positive control group,even though the mi cronucleus rate of the low dose group of DPPD(62.5 mg/kg) combined with cyclophosphamide didn ’t change obviously,the micronucleus rates of the middle and high dose groups of DPPD(250、1000 mg/kg) combined with cyclophosphamide increased obviously(P 0.01),and the inhibition rates of bone marrow micronucleus were 35.58% and 61.54% re spectively.Conclusion DPPD shows no impact on bone marrow micronucleus after singly administering DPPD orally thirty days,but DPPD can antagonize the bone marrow micronucleus inducing effect of cyclophosphamide in mice.
