实验大鼠胰岛分离方法标准化研究

摘　　要：目的借鉴成人胰岛分离方法,采用商品化的试剂,建立较为稳定的实验大鼠胰岛分离方法。方法分别用传统方法和标准方法各消化12只大鼠的胰腺,比较消化后和纯化后的胰岛得率,以胰岛素释放试验比较两组胰岛的功能。对照组与标准组胰岛得率、活率和功能指标的比较,采用独立t检验分析。结果标准组消化后和纯化后胰岛数量为(891±53)个和(590±88)个,对照组为(888±115)个和(562±105)个(t=0.082,0.708;P=0.935,0.486);胰岛素释放指数为2.57±0.17和2.50±0.52,(t=0.443,P=0.662)差异无统计学意义。结论标准化的胰岛分离方法,可减少实验误差、利于尽快掌握分离技术,保证胰岛的得率和功能。特别是进行胰岛移植实验时,标准化的胰岛分离方案可提高移植效果,保证实验的可靠性。To establish a relatively stable method for islet isolation from rats by using similar isolation method for human adult islet. Methods Islets were digested according to traditional method and standard method respectively in 12 rats. The yield of purified islets and their function which was evaluated by insulin releasing tests were compared between the two groups. Results The yields of islet after digestion and purification in standard group were 891 ± 53 islets per pancreas and 590 ±88 islets per pancreas respectively, which were higher than the traditional group （888±115 and 562± 105, respectively）. No significant difference was observed （t = 0.082, 0.708. P = 0.935, 0.486） . The islet function shown in insulin releasing tests was similar between the two groups （2.57 ± 0.17 vs 2.50 ± 0.52 respectively）. Conclusions The standard islet isolation method greatly reduces the experimental error and can be learned more quickly. The standard islet isolation method could improve the transplantation efficiency.
