人脐带间充质干细胞在胶原支架材料体外复合培养的生物学特性研究

摘  要:目的:观察体外人脐带间充质干细胞(mesenchymal stem cells,MSCs)黏附胶原蛋白支架条件下的干细胞免疫表型及成骨特性的变化,为复合载体移植提供实验依据。方法:将人脐带间充质干细胞静态接种于胶原蛋白支架上,经体外培养两周后,检测细胞在胶原蛋白材料中的干细胞CD44、CD105、HLA-DR、Ⅰ型胶原表达,观察细胞特性变化。结果:细胞在支架上有较好的粘附,CD 44、CD105、Ⅰ型胶原表达阳性,HLA-DR表达阴性。结论:混合培养中人脐带MSCs不仅与胶原蛋白有较好的组织相容性,且细胞在支架上培养仍保持了其干细胞的特点,Ⅰ型胶原表达阳性,提示具有潜在的成骨能力,为人脐带间充质干细胞组织工程体外预制提供实验基础。Objective: To research the biological properties of human umbilical mesenchymal stem cells ( MSCs I cultured with collagen scaffolds in vitro, provide experimental basis for application. Method: After 2 weeks of culture with collagen scaffold, immune and histochemical morphology of the cells on the collagen scaffold were observed. Result: The UC-MSCs immunohistochemical staining revealed that the cells were positive for CD44, CD105, type I collagen. And negative for HLA-DR. Conclusion: Collagen scaffold has favorable compatibility with human UC-MSCs. The cells cultured with collagen scaffold can keep the properties of stem cells and have the potential to produce bone.
