摘  要:目的探讨生脉散对急性肝衰竭内毒素血症模型大鼠的作用机制。方法选取50只SD大鼠,随机分为正常对照组、肝衰竭模型组(简称肝衰组)、肝衰竭模型加生脉散治疗组(简称肝衰+中药组)、肝衰竭内毒素血症模型组(简称LPS组)及肝衰竭内毒素血症模型加生脉散组(简称LPS+中药组),每组各10只。采用D-氨基半乳糖腹腔注射建立大鼠急性肝衰竭模型,在此基础上以大肠杆菌脂多糖(LPS)腹腔注射建立肝衰竭内毒素血症模型。肝衰+中药组与LPS+中药组造模前1h灌服生脉饮干预治疗。分别于造模2、8h后观察比较5组血清白细胞介素(IL)-6、IL-1β、肿瘤坏死因子-α(TNF-α)及LPS水平变化。结果肝衰组及LPS组造模2h后血清IL-6、IL-1β水平变化与正常对照组比较差异有统计学意义(P<0.05),LPS组血清LPS及TNF-α水平变化与正常对照组比较差异有统计学意义(P<0.05);肝衰+中药组造模2h后血清IL-1β水平变化与肝衰组比较差异有统计学意义(P<0.05),LPS组血清IL-6、IL-1β、TNF-α及LPS水平变化与肝衰组比较差异有统计学意义(P<0.05);LPS+中药组造模2h后血清IL-6、IL-1β、TNF-α及LPS水平变化与LPS组比较差异有统计学意义(P<0.05)。肝衰组及LPS组造模8h后血清IL-6、IL-1β及LPS水平变化与正常对照组比较差异有统计学意义(P<0.05);肝衰+中药组造模8h后血清IL-1β水平变化与肝衰组比较差异有统计学意义(P<0.05),LPS组血清IL-6、IL-1β及LPS水平变化与肝衰组比较差异有统计学意义(P<0.05);LPS+中药组造模8h后血清IL-1β及LPS水平变化与LPS组比较差异有统计学意义(P<0.05)。结论生脉散能调节急性肝衰竭内毒素血症模型大鼠血清TNF-α、IL-6、IL-1β含量变化,降低内毒素水平,不仅可减少炎性介质的产生,而且也阻断了炎性介质及内毒素本身对机体的损伤。Objective To study the mechanism of Shengmai powder on inhibition of endotoxemia in rats with acute liver failure.Methods50 SD rats were randomly divided into normal control group(group A),liver failure model group(group B),liver failure model treatment group(group C),liver failure with endotoxemia model group(group D) and liver failure with endotoxemia model treatment group(group E).Rat model of acute liver failure were established using intraperitoneal injection of D-galactosamine,liver failure endotoxemia model were established by intraperitoneal injection of E.coli lipopolysaccharide(LPS).Rats in group C and E received orally intervention treatment of Shengmai powder at 1 h before modeling.The levels of TNF-α,IL-6,Il-1βand LPS were observed at 2,8 h after intervention in five groups.Results There was significant difference between group B,D and A on the levels of IL-6 and Il-1βin serum at 2 h after making model(P 〈0.05).There was significant difference between group D and A on the levels of LPS and TNF-αin serum(P 〈0.05).There was significant difference between group B and C on the level of Il-1βin serum at 2 h after making model(P 〈0.05).There were significant differences on the changes of levels of TNF-α,IL-6,Il 1βand LPS in serum of group D compared with those in ser-um of group B(P 〈0.05).There were significant differences between group D and E on the changes of levels of TNF-α,IL-6,Il-1βand LPS in serum at 2 h after making model(P 〈0.05).There was significant difference between group B,D and A on the levels of IL-6,Il-1βand LPS in serum at 8 h after making model(P 〈0.05).There was significant difference between group B and C on the level of Il-1βin serum at 8 h after making model(P0.05).There was significant difference between group D and E on the changes of levels of LPS and Il-1βin serum at 8 h after making model(P 〈0.05).Conclusion Shengmai powder can regulate the levels of TNF-α,IL-6 and Il-1βof endotoxemia rat with a
