摘  要:目的:探讨白英生物碱(STA)诱导肺腺癌A549细胞凋亡及对NF-κB信号通路相关基因表达的影响。方法:采用STA处理体外培养的A549细胞,用MTT法检测细胞增殖抑制效果,Annexin V-FITC/PI双染后用流式细胞仪检测细胞凋亡率,Western blot检测细胞核NF-κB/p65和胞浆Survivin、IκBα、p-IκBα蛋白表达。结果:STA对A549细胞增殖具有较强的抑制作用,并呈剂量-时间依赖性;STA作用24 h后,细胞凋亡率显著上升,IκBα表达显著上升,NF-κB/p65、Survivin和p-IκBα表达显著下降。结论:白英生物碱可明显诱导A549细胞凋亡,其机制可能与抑制NF-κB/p65信号通路有关。Objective:To investigate the effects of solanum lyratum Thunberg alkaloid(STA) on induction of apoptosis and the ex- pression of NF-KB signaling pathway related genes in A549 cells. Methods:AS49 cells was treated with STA in vitro. The proliferation inhibitory effect was evaluated by MTT assay. Induction of cell apoptotic rate was determined by flow cytometry method (FCM) after An-nexin V-FITC/PI double staining. The expression of NF-κB/p65 in nuclei, Survivin, IκBα and p-IκBα in cytosol were detected by west-ern blot. Results:STA exhibited strong proliferation inhibitory effect in a dose-and -time-dependent manner against A549 cells. After treated with STA for 24 h, the apoptotic rate was increased significantly. The expression of IκBα protein was increased markedly, while those of NF-κB/p65, Survivin and p-IκBα proteins were decreased markedly. Conclusion : STA can induce apoptosis of lung adenoearci-noma A549 cells,its mechanisms may be related to inhibition of NF-κB signaling pathway.
