摘  要:目的研究Sirtuin6(SIRT6)在血管紧张素Ⅱ(AngⅡ)诱导的新生大鼠心肌细胞肥大中的表达变化情况。方法在AngⅡ诱导的心肌细胞肥大模型中采用实时定量RT-PCR(Real Time PCR)筛选了Sirtuin家族各亚型的mRNA水平变化,并进一步通过Real TimePCR和Western Blot检测AngⅡ诱导SIRT6的mRNA和蛋白水平变化。结果 AngⅡ诱导SIRT6表达上调呈现剂量和时间依赖性。100 mmol/L AngⅡ作用于心肌细胞24 h,SIRT6的mRNA和蛋白水平分别为空白对照组的3.31±0.17倍和4.37±0.21倍。结论心肌肥大体外模型中,SIRT6的mRNA和蛋白水平均明显升高。Objective To measure the expression of Sirtuin 6 (SIRT6) in Angiotensin II cardiomyocyte hypertrophy model. Methods Quantitative RT-PCR was performed to screen the ( Ang effect 11 ) induced of Ang II on mRNA expressions of all 7 sirtuins in primary neonatal rat cardiomyocytes. The SIRT6 mRNA and protein levels were examined by quantitative RT-PCR and Western blot. Results The SIRT6 mRNA and protein expression levels were elevated in a dose- and time-dependent manner. After treated with 100 mmoL/L of Ang I/ for 24 h, the mRNA and protein levels of SIRT6 were increased by 3.31 ±0. 17 and 4.37 -+0.21 fold, respectively, compared with untreatment group. Conclusion The mRNA and protein levels of SIRT6 are elevated in in vitro model of cardiac hypertrophy.
