摘 要:目的探讨VEGF-165-GMs复合自体颗粒骨修复兔陈旧性桡骨缺损的效果。方法 24只新西兰大白兔双侧桡骨均建立1.5 cm的陈旧性骨缺损实验模型,随机分为A、B、C三组,A组植入自体颗粒骨+VEGF-165-GMs 1mg;B组植入200 mg自体颗粒骨;C组VEGF-165-GMs 1 mg;分别于术后3、6、9周切取标本,通过X线摄片、骨密度测量、生物力学测试、组织形态学观察等手段,观察各个时期新骨形成的情况。结果 A、B组均能较好地修复陈旧性骨缺损,A组术后各时间点X线评分、骨密度测量、生物力学测试、组织形态学观察均明显优于B组。C组各时间点均无骨性愈合迹象,9周时可见骨断端完全硬化,髓腔封闭。结论 VEGF-165-GMs复合自体颗粒骨修复陈旧性骨缺损可以明显促进局部血运重建,促进新生骨形成,提高骨改建塑形的速度和质量。Objective To explore the effect about composite of VEGF-165-GMs and autogenous morselized bone on the repairing obsolete radial defects in rabbits. Methods Make obsolete radial defect model on 24 rabbits’double radial,then randomly divided into A, B, C groups. A group treated with the compound of the VEGF-165-GMs and autogenous morselized bone on obsolete radial defects, B group treated with autogenous morselized bone only, C group treated with VEGF-165-GMs ; animals were put to death at different time points(3,6,9 week) to get the model, through X-ray imaging, bone density measure, biomechanics test, histology compare to understand the growth condition of new bone at different time points. Results A,B groups both active repair obsolete radial defects in rabbits.But A group reflected better in X-ray imaging, bone density measure ,biomechanics test, histology compare. C group has no synostosis at any time. In 9 week the broken ends of fractured bone to become complete sclerosis, and the canals of de Candolle were blockaged. Conclusion The composite of VEGF-165-GMs and autogenous morselized bone can distinguished enhance local revascularization, and promote neoplastic bone formation, improve the speed and quality of bone remodeling and bone moulding.
