摘 要:目的观察外周血白细胞(WBC)中HM74A表达与2型糖尿病(T2DM)的相关性,并探讨HM74A信号对炎症因子的调节作用。方法收集健康人(对照组)与T2DM患者静脉血,提取mRNA,利用半定量逆转录聚合酶链式反应(RT-PCR),检测WBC中HM74A、核因子-κB(NF-κB)、白细胞介素(IL)-1基因表达水平,分析对照组与T2DM组HM74A及其下游因子NF-κB、IL-1βmRNA表达的差异性。结果 RT-PCR结果显示,T2DM组HM74A、NF-κB、IL-1β基因表达水平显著高于对照组(P<0.05)。相关性分析结果显示WBC中HM74A与其下游因子NF-κB、IL-1β的表达量呈正相关(r=0.402,r=0.451,均P<0.05);HM74A、NF-κB与血糖呈正相关(r=0.179,r=0.358,均P<0.05)。结论 T2DM患者外周血WBC中HM74A及炎症因子NF-κB、IL-1βmR-NA表达量显著增高,提示HM74A信号可能参与T2DM的发生和发展,且对炎症因子有一定的调节作用。Objective To investigate the association between HM74A expression in the peripheral blood leukocytes(PBLs) and type 2 diabetes(T2DM),and to study the regulation of inflammatory factors by HM74A signaling.Methods Peripheral blood samples and biochemical data were collected from healthy individuals(normal controls) and T2DM patients.The semi-quantitative reverse transcription polymerase chain reaction(RT-PCR) was used to measure mRNA levels of HM74A,NF-κB,and IL-1β in PBLs.Results According to RT-PCR,the mRNA levels of HM74A,NF-κB,and IL-1β in T2DM group were significantly higher than those in control group(P〈0.05).Correlation analysis showed that there was significantly positive correlation between HM74A and NF-κB(r = 0.402,P〈0.05),IL-1β(r = 0.451,P〈0.05),or fasting plasma glucose(FPG;r = 0.179,P〈0.05);so was the significantly positive correlation between NF-κB and FPG(r = 0.358,P〈0.05).Conclusion HM74A signaling may be associated with T2DM,and plays a role in regulation of the inflammatory cytokines.
