摘  要:目的探讨海风藤对急性肺损伤(ALI)新生大鼠水通道蛋白5(AQP5)的影响。方法 150只新生健康SD大鼠腹腔注射内毒素(LPS)5 mg/kg后随机分为对照组和海风藤低浓度组、海风藤高浓度组,对照组注射LPS后不行治疗,海风藤低浓度、高浓度组在未到处死取材时间分别按10 mg/kg和30 mg/kg海风藤提取液雾化,雾化时间为注射LPS后2、26、50、74、98、122及146 h。按处死时间分为5个亚组:8、24、48、72 h及7 d组,收集肺组织予病理学检查及测肺湿/干重比值(W/D),用免疫印迹法测肺组织AQP5的表达。结果 (1)肉眼观察予LPS 48 h后同一时间点海风藤高浓度组均明显较对照组及低浓度组水肿程度轻,出血肺叶面积小。光镜下对照组及海风藤低浓度组可见大量红细胞,海风藤高浓度组为少量红细胞渗出。(2)对照组及海风藤低、高浓度组W/D比值均升高,海风藤高浓度组在24 h~7 d亚组与对照组比较差异均有统计学意义(P<0.01)。(3)AQP5表达海风藤高浓度组与对照组在24 h至7 d各亚组差异均有统计学意义(P<0.01),海风藤低浓度组与对照组比较仅在72 h、7 d亚组差异有统计学意义(P<0.05)。结论雾化吸入海风藤可减轻ALI新生鼠的肺水肿及出血程度,海风藤抗炎作用能减少Ⅰ型肺泡上皮细胞受损,并促进水肿吸收。高浓度海风藤治疗效果优于低浓度。Objective To evaluate the effects of Caulis Piperis Kadsurae on Aquaporin 5(AQP5) in neonatal rats with acute lung injury (ALI).Methods Lipopolysaccharides (LPS) was injected in 150 healthy neonatal SD rats, which were randomly divided into untreated group (Group C) ,low Caulis Piperis Kadsurae treatment group (Group L) and high Caulis Piperis Kadsurae treatment group (Group H).According to the timing of sacrifice the subjects were further divided in to subgroups.The lung tissues were collected for pathologic observation and measurement of lung wet/dry weight ratio (W/D).The expression of AQP5 was assessed by Western blot.Results Severer edema and pulmonary hemorrhage were observed in Group L when comparing with those in Group H 48 hours after LPS induction.Obvious red blood cell aggregation was observed in Group C and Group L,while it was significantly alleviated in Group H.Although significant elevation of W/D was revealed in all the 3 groups,there were significant difference between Group C and H 24 hours after induction (P〈0.01).There were significant differences in AQP5 expression between Group H and Group C 24 hours after induction,while these significant differences were only revealed between Group L and Group C 3 and 7 days after induction (P〈0.05).Conclusion Atomizing inhalation of Caulis Piperis Kadsurae can reduce ALI of neonatal rats with pulmonary edema and pulmonary hemorrhage via the anti - inflammatory effect on type Ⅰ alveolar epithelial cells, and the promotion of edema absorption.High concentration of Caulis Piperis Kadsurae treatment is better than low concentration.
