摘  要:目的观察三七总甙(PNS)对糖尿病皮肤溃疡(DSU)大鼠单核细胞趋化蛋白-1(MCP-1)的影响。方法选择成年Wistar大鼠50只,随机分为对照组、DSU模型组和PNS高、中、低剂量处理组,每组10只。放射免疫法检测大鼠血清MCP-1水平,反转录-聚合酶链反应、Western blotting检测大鼠溃疡处组织中MCP-1的表达。结果与对照组比较,模型组大鼠血清MCP-1水平、溃疡处组织MCP-1 mRNA、蛋白水平显著升高(P<0.05);而PNS高、中、低剂量处理组血清MCP-1水平及溃疡处组织MCP-1 mRNA及蛋白水平均较模型组下降(P<0.05),且PNS高、中剂量处理组显著低于低剂量处理组(P<0.05),高剂量处理组显著低于中剂量处理组(P<0.05)。结论 PNS可降低MCP-1的表达,有效防治DSU的发生发展。Objective To investigate the effects of panax notoginsenoside(PNS) on monocyte chemotactic protein-1(MCP-1) in diabetic skin ulceration(DSU) rats.Methods Fifty adult Wistar rats were randomly divided into control group,model group,PNS high-dose group,PNS middle-dose group and PNS low-dose group,ten rats in each group.The contents of MCP-1 in serum were detected by radioimmunity,and the expression of MCP-1 in ulcer tissue was detected by reverse transcription polymerase chain reaction(RT-PCR) and Western blotting.Results The MCP-1 levels in serum,and the MCP-1 mRNA and protein levels in ulcer tissue of model group were significantly higher than those of control group(P0.05).The MCP-1 levels in serum,and the MCP-1 mRNA and protein levels in ulcer tissue of PNS high-dose group,PNS middle-dose group and PNS low-dose group were significantly lower than those of model group(P0.05);them in PNS high-dose group and PNS middle-dose group were lower than PNS low-dose group(P0.05),and in PNS high-dose group were lower than PNS low-dose group(P0.05).Conclusion PNS can decrease the expression of MCP-1 and prevent the development of DSU.
