摘  要:目的:观察心复康口服液对心肌梗死心衰大鼠心肌组织肌型肌酸激酶同工酶(CK-MM)mRNA及CK-MM蛋白表达的影响。方法:采用结扎SD大鼠左冠状动脉前降支复制心肌梗死后心衰大鼠模型,大鼠随机分为假手术组(Sham组)、模型组(Model组)、Capt 0.1 g.kg-1组(Capt组)、心复康口服液组5.4 g.kg-1(XFK组),分别于术后24 h灌胃给药,给药4周末,用Real-time RT-PCR方法,Western blotting方法检测心肌细胞能量穿梭关键酶CK-MM mRNA和蛋白表达水平。结果:①与假手术组比较,模型组左心室心肌CK-MM的mRNA表达水平下调,在第4周末,统计学有显著性意义(P<0.01);与模型组比较,Capt组、XFK组心肌CK-MM mRNA、蛋白表达均上调,在第4周末,统计学有显著性意义(P<0.01)。②与Captopril组比较,XFK组心肌CK-MM mRNA表达水平在第4周末下调,统计学有显著性意义(P<0.05);XFK组心肌CK-MM蛋白表达水平在第4周末比较无显著性意义。结论:心复康口服液可上调心梗后心衰大鼠心肌细胞CK-MM mRNA的表达,促进CK-MM蛋白含量增加,达到改善CHF心肌细胞能量穿梭紊乱,重塑心梗后心衰大鼠心肌能量代谢作用。Objective: To study the effect of Xinfukang (XFK) oral liquid on MM isoenzyme of creatine kinase (CK-MM) mRNA and CK-MM protein expression level in rat of heart failure model after myocardial infarction. Method: The rat heart failure mode after myocardial infarction was created by ligating the left anterior descending coronary arteries. All the rats were randomly divided into 4 groups: sham group, model group, XFK 5.4 g·kg^-1 group and captopril 0. 1 g·kg^-1 group. After 24 hours of modeling, rats were ig given XFK and captopril for 4 weeks, Cardiac function, myocardial uhrastructure and key enzymes of myocardial energy shuttle, CK-MM mRNA and protein expression level were separately measured in all groups. Result: (~)Compared with model group, in captopril group and XFK group CK-MM mRNA and protein expression increased (P 〈 0.01 ). (~) Compared with captopril group, in XFK group CK-MM mRNA expression level decreased (P 〈 0.05). In XFK group CK-MM protein expression has no significant change. Conclusion: XFK oral liquid can effectively increase the expression level of CK-MM mRNA, as well as the protein content of CK-MM to improve myocardial energy shuttle and remodel the process of myocardial energy metabolism function.
