摘 要:成功构建了能够高效表达鲎素TPⅠ基因的组成型毕赤酵母表达菌株,发酵培养后上清对分属于13个属的61株菌抑菌活性良好。应用肉汤微量稀释法测定发酵上清对金黄色葡萄球菌的抗菌活性,用扫描电镜观察其对金黄色葡萄球菌的抑制作用,并探讨发酵上清对小鼠皮肤感染模型的治疗作用。结果显示,基因工程鲎素对金黄色葡萄球菌最小抑菌浓度(MIC)为(3.46±0.76)mg/L,亚抑菌浓度为(0.865±0.22)mg/L。基因工程鲎素对金黄色葡萄球菌小鼠感染模型的治疗效果与抗生素差异不显著(P>0.05),且具有刺激小鼠血管内皮生长因子表达水平增加,加速血管生成和血管发生,促进皮肤创伤愈合的功能;表明基因工程鲎素对金黄色葡萄球菌感染小鼠具有良好的治疗效果。The expression of tachyplesin Ⅰ (TP Ⅰ )Pichia pastoris has been successfully estab-lished in this study. Genetic engineering TP I displayed broad-spectrum antibacterial activity a- gainst 13 genus 61 strains bacteria. A broth micro-dilution method was used to determine the anti-microbial activity of genetic engineering tachyplesin against Staphylococcus aureus, then the de-pressant effect of it on Staphylococcus aureus was observed by scanning electron microscopy,and it's therapeutic effect was evaluated in murine model of infetion. The results showed that its minimum in-hibitory concentration (MIC) was (3.46 ± 0.76) mg/L, sub-MIC was (0. 865±0.22) mg/L. There was no significant difference of the therapeutic effect between the genetic engineering tachYplesin and antibiotic group in Staphylococcus aureus infection model of mouse(P〉0.05). The genetic en-gineering tachyplesin could stimulate the growth factor levels of vascular endothelial,could accel- erate growth of new angiogenesis,and promote skin wound healing. The genetic engineering tachy- plesin have a therapeutic effect in a mouse skin infection model.
