大承气汤对PCAS兔心肺组织蛋白酶激活受体-1表达的影响

摘  要:目的探讨大承气汤对心脏骤停后综合征(PCAS)兔心肺组织蛋白酶激活受体-1(PAR-1)表达的影响。方法将20只兔用来制备PCAS模型,另外10只作为正常对照组。采用窒息性心脏骤停制备兔PCAS,窒息性心脏骤停采用气管夹闭诱导。18只复苏成功兔随机分为PCAS组和大承气汤组。大承气汤组在自主循环恢复(ROSC)后15min给予大承气汤16g/kg灌胃;其他两组同时间点灌胃等量生理盐水。各组48h后处死动物,取心肺组织,采用相对定量实时逆转录-聚合酶链反应(RT—PCR)测定PAR-1mRNA表达。结果与正常对照组比较,PCAS组兔心肺组织中PAR-1mRNA表达较正常对照组显著升高(均P〈0.01)。大承气汤干预后心肺组织中PAR-1mRNA表达均较PCAS模型组显著下降(P〈0.01或P〈0.001)。结论大承气汤对PCAS兔心肺组织PAR-1表达有抑制作用,这可能是大承气汤治疗多器官功能障碍综合征(MODS)的机制之一。Objective To investigate the influence of Dachengqi granule on the expression of proteinase - activated receptor - 1 ( PAR - 1 ) mRNA in the heart and lung of rabbits with post - cardiac arrest syndrome (PCAS). Methods Twenty healthy male Japanese rabbits were used to establish PCAS model, and another ten rabbits as normal control group. The model of PCAS was established by asphyxia- induced cardiac arrest which was induced by tracheal tube clamping. Eighteen of twenty rabbits had return of spontaneous circulation ( ROSC), and were randomly assigned to two study groups, PCAS group and vehicle control. Dachengqi granule group: 15 g/kg Dachengqi granule was given by intra -gastric administration and twice a day 15 min after ROSC. In the other two groups, intra- gastric administration with same amount of saline was given at the same time. After 48 hours, animals in each group were sacrificed, and heart and lung samples were taken and analyzed by relatively quantitative real - time RT - PCR method to show the expression of PAR - 1 mRNA. Results Expressions of PAR - 1 mRNA in heart and lung in PCAS group were higher than those in normal control group (P 〈 0. 01 ). After intervention by Dachengqi granule, the expressions of PAR - 1 mRNA were lower than those in PCAS group ( P 〈 0.01 or P 〈 0.001 ). Conclusion Dachengqi granule inhibits the expression of PAR - 1 in heart and lung in rabbits with PCAS. It may be one of the mechanisms by which Dachengqi granule treat multil31e organ dysfunction syndrome (MODS).
