摘　　要：目的:制备菲牛蛭活性组分。方法:经微滤及PEG-pH沉淀,除去菲牛蛭匀浆液中的杂质,再通过冷冻干燥法制备菲牛蛭活性组分,并考察该组分对APTT、TT、PT值的影响,初步评价其抗凝血活性。结果:所制备的菲牛蛭活性组分冻干粉,复溶性好,抗凝血酶活性为324.52 ATU.g-1,比活性为2 480.16 ATU.mg-1,总活性回收率为79.58%,且能显著延长APTT、TT、PT值。结论:以本方法所制备的菲牛蛭活性组分,活性回收率高,制备方法简单可靠,重复性好。OBJECTIVE To prepare the active constituents of Poecilobdella manillensis Lesson with highly activity.METHODS Firstly,the impurities from the Poecilobdella manillensis homogenate could be removed by microfiltration and PEG precipitation.Then the active constituents could be prepared by freeze drying.And its anticoagulant activities in vitro were appreciated preliminarily by the method of APTT,PT and TT test.RESULTS The freeze-dried powder of the active constituents of Poecilobdella manillensis which could extend the APTT,TT and PT,got a good appearance and could be reconstruction well.Furthermore the antithrombotic activity was 324.52 ATU·g^-1the specific activity of obtained products reached 2480.16 ATU·mg-1 and the recoveries of ATU reached 79.58%.CONCLUSION The method used to prepare freeze-dried powder of active constituents of Poecilobdella manillensis was simple and convenient,with high active recovery rate and good reproducibility.
