摘  要:目的探讨氢饱和生理盐水对急性CO中毒大鼠脑损伤的保护作用。方法 42只SD大鼠随机分为对照组、CO染毒组和氢饱和生理盐水治疗组。通过Morris水迷宫实验观察大鼠脑功能变化,尼氏染色观察大鼠脑组织神经元死亡率,电镜观察神经元超微结构变化。结果 Morris水迷宫定向航行实验,CO组逃避潜伏期较对照组显著延长(P﹤0.05),H2组大鼠逃避潜伏期较染毒组缩短。空间探索实验CO组第一象限游泳时间较对照组缩短(P﹤0.05),H2组游泳时间较染毒组延长。尼氏染色标本上,对照组脑内神经元形态正常,死亡细胞少;染毒组脑内出现大量坏死细胞,氢饱和生理盐水组坏死细胞较CO组明显减少(P﹤0.05)。电镜观察发现对照组神经元形态正常,染毒组出现缺血性改变,而氢饱和生理盐水组细胞形态基本正常。结论 H2饱和生理盐水可改善由CO中毒所致的行为学异常;可降低脑组织神经元的死亡率;减轻神经元缺血性改变的程度。提示H2饱和生理盐水对急性CO中毒脑损伤具有一定的保护作用。OBJECTIVE To observe the protective effects of hydrogen rich saline on rats with acute carbon monoxide (CO) poisoning. METHODS 42 Sprague-Dawley (SD) male rats were divided into three groups including control group, CO group and H2 rich saline group. Morris water maze experiments were monitored for cognitive function. Death rate of neurons was assessed by Nissl's staining. Ultramicrostructure of neurons was observed by electron microscope. RESULTS Compared with control group, the escape latency was longer (P 〈 0.05) and the swimming time in I quadrant (P 〈 0.05) was shorter in CO group. However, H2 rich saline treatment made the escape latency shorter (P 〈 0.05) and the swimming time in I quadrant longer (P 〈 0.05) than CO group. The ultramicrostructure of neurons stayed normal and the death rate was small in control group. The death rate increased in the CO group and the rate in H2 rich saline group decreased obviously more than CO group (P 〈 0.05). The ultramicrostructure observed in the control group stayed normal, the ischemic change occurred in the CO group and the nhramicrostructure stayed normal in H2 rich saline group. CONCLUSION H2 rich saline treatment improves the cognitive function, decreases the death rate of neurons and lessenes neuron ultramierostructure changes after acute CO poisoning. It suggests that H2 rich saline is effective to acute CO poisoning.
