PD98059对体外CCl_4刺激的肝星状细胞增殖及I型胶原纤维表达的影响

摘　　要：目的探讨特异性MEK抑制剂PD98059对体外肝星状细胞增殖及I型胶原纤维表达的影响。方法将液氮保存下的肝星状细胞株,于37℃、5%CO2孵育箱中复苏传代培养,同步化后将细胞分为3组:对照组、CCl4组、CCl4+不同剂量PD98059(20、50、100μmol/L),用透射电镜观察细胞超微结构,MTT比色法检测细胞增殖情况及免疫细胞化学法测定I型胶原纤维表达。结果透射电子显微镜可见PD98059组比CCl4组凋亡细胞显著增多,细胞数目减少;CCl4刺激HSC后,细胞明显增殖(P﹤0.05),各剂量组PD98059对细胞增殖均有抑制作用(P﹤0.05),抑制率分别为21.84%、38.27%、46.65%;CCl4组HSC中的I型胶原表达与对照组比较显著增加(P﹤0.05),PD98059明显抑制I型胶原表达(P﹤0.05)。结论 PD98059对CCl4刺激HSC增殖及I型胶原的表达具有抑制作用。OBJECTIVE To find out the effect of specific MEK inhibitor on proliferation and type I collagen expression of hepatic stellate cell （HSC） stimulated by carbon tetrachloride （CCL） in vitro. METHODS Hepatic stellate cell lines incu- bated in 37℃, 5% carbon dioxide incubation box were divided into three groups： the control group, carbon tetraehloride （CCL） group and CCL plus different dose PD98059 （20, 50, 100mol/L） groups. The uhrastructure was observed with transmission electron microscope, cell proliferation was assessed by MTr colorimetric assay and type I collagen fiber expres- sion was determined by immunocytoehemistry method. RESULTS Transmission electron microscopy showed that apoptotic cells in PD98059 group were increased obviously, the number was less than that in CCI4 group and the number of cells was reduced after the inhibitor PD98058 treatment. After the HSC was stimulated by CCL, cells proliferated obviously （P 〈 0.05）, different doses of PD98059 had inhibitory effects on the cell proliferation （P 〈 0.05） and the inhibition rates were 21.84%, 38.27% and 46.65% respectively. Compared with the control group, expression of type I collagen in CC14 group increased ob- viously （P 〈 0.05）, while PD98059 inhibited the expression of type I collagen （P 〈 0.05）. CONCLUSION PD98059 has in- hibition effect on the proliferation and expression of type I collagen in hepatic stellate cell stimulated by CCL.
