摘 要:目的观察莪术油对人卵巢癌SKOV3细胞增殖和细胞周期的影响及其铂类耐药细胞SKOV3/DPP的逆转作用。方法噻唑蓝比色法观察不同质量浓度莪术油对SKOV3细胞增殖抑制作用,计算半数抑制浓度;流式细胞技术(FCM)检测莪术油对SKOV3细胞周期的影响;2×2析因试验分析顺铂联合莪术油对SKOV3/DDP的交互作用,以及增加敏感的作用。结果莪术油对SKOV3细胞的增殖有明显的抑制作用,半数抑制浓度为34μg/mL;FCM结果显示莪术油组较对照组细胞G1期比例明显升高,而G2、S期显著下降(P<0.01);析因实验结果表明顺铂和莪术油之间存在交互作用,莪术油可以增强顺铂对SKOV3/DDP细胞的抑制作用。结论莪术油对人卵巢癌SKOV3有显著抑制作用,可将其细胞周期阻滞于G0/G1期;莪术油可以增加人卵巢癌SKOV3/DDP细胞对顺铂的敏感性。AIM To observe the effect of zedoary oil on proliferation and the cell cycle in the human ovarian cell line SKOV3 and the reversal effect of zedoary oil on cisplatin-resistant ovarian cancer cell line SKOV3/DDP.METHODS The SKOV3 cells were treated by different concentrations of zedoary oil and MTT was used to detect the IC50 rates of proliferation inhibition.Flow cytometry(FCM) was used to examine the influence of zedoary oil on the cell cycles of SKOV3 cells.2×2 factorial designs were used to analyze the interaction of cisplatin and zedoary oil to SKOV3/DDP cell and sensitization to cisplatin resistance.RESULTS Zedoary oil inhibited the proliferation of SKOV3 cells remarkably and the inhibition rate increased with the increase of the concentration of zedoary oil,and IC50 was 34 μg/mL.FCM showed that the portion of G1 phase in zedoary oil group increased and portion of both G2 and S phase decreased(P0.01).2×2 factorial designs trial showed that there existed interaction effect between the cisplatin and zedoary oil.The latter enhanced sensitivity of cisplatin to the SKOV3/DDP cell.CONCLUTIONS The experiment reveals that zedoary oil could inhibit the proliferation of SKOV3 cells remarkably and arrest at the G0/G1 phase and improve the sensibility of SKOV3/DDP cell to cisplatin.
