摘  要:目的观察香青兰总黄酮预处理对大鼠心肌缺血再灌注损伤的保护作用及可能的机制。方法采用结扎大鼠左冠状动脉前降支的方法建立大鼠心肌缺血再灌注损伤模型。记录心电图S-T段变化,观察心肌组织病理学改变,测定各组血清乳酸脱氢酶(LDH)、肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)、谷草转氨酶(AST)、总超氧化物歧化酶(T-SOD)、谷胱甘肽过氧化物酶(GSH-Px)和髓过氧化物酶(MPO)的活性及血清中丙二醛(MDA)、白细胞介素-1(IL-1)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)的水平。结果与模型组比较,香青兰总黄酮能抑制缺血再灌注时心电图S-T段的抬高及缺血心肌病理学改变,降低血清中LDH、CK、CK-MB、AST和MPO的活性及MDA、IL-1、IL-6和TNF-α的水平,并能够提高血清T-SOD和GSH-Px的活性。结论香青兰总黄酮对心肌缺血再灌注损伤心肌具有明显的保护作用,这种作用可能与抗氧化损伤和抑制炎症反应机制有关。AIM To investigate the protective effect and possible mechanism of total flavones from Dracocephalum moldavica pretreatment on myocardial ischemia-reperfusion injury in rats.METHODS Myocardial ischemia-reperfusion model was established on rats by ligation of coronary artery.S-T segment change was recorded and the histopathological changes were observed.The activities of lactate dehydrogenase(LDH),creatine kinase(CK),creatine kinase isoenzyme(CK-MB),acetic transaminase(AST),total superoxide dismutase(T-SOD),glutathione peroxidase(GSH-Px),myeloperoxidase(MPO) and the level of malonaldehyde(MDA),nterleukins-1(IL-1),nterleukins(IL-6),tumor necrosis factor-α(TNF-α) in serum were detected.RESULTS Compared with model group,the experiment revealed that total flavones from Dracocephalum moldavica attenuated S-T segment elevation and myocardium pathological changes decreased the activities of LDH,CK,CK-MB,AST,MPO and the level of MDA,IL-1,IL-6,TNF-α in serum,and improved the activities of T-SOD and GSH-Px.CONCLUSION The experiment shows that total flavones from Dracocephalum moldavica present protective effects against ischemia-reperfusion injury in rats,and its mechanism is probably related to resist oxidation damage and inflammatory reaction.
