摘 要:目的初步评价四川省川牛膝居群的遗传多样性。方法应用ISSR分子标记对川牛膝主产地6个居群,23份川牛膝样本进行了植物性状研究及遗传性研究。结果经过引物筛选试验选出4个呈现多态性的引物,总共扩增条带数为43条。POPGENE分析结果表明,川牛膝的遗传变异在物种水平上,基因多样性指数(He)为0.189 2,多态位点百分率(PPL)为67.4%,Shannon信息指数(Ho)为0.322 4;在居群水平上,He为0.717,PPL为49.76%,Ho为0.250 9。居群遗传距离的变化范围分别为0.52~1.00。结论四川主产地川牛膝物种间遗传多样性较大,居群间遗传多样性较小,遗传变异主要由异花授粉造成。AIM To preliminarily evaluate the genetic diversity of Cyathula officinalis Kuan populations planted in Sichuan Province.METHODS ISSR molecular markers were performed on 23 samples of Cyathula officinalis Kuan collected from six populations for the study of plant traits and genetic research. RESULTS Four polymorphic ISSR primers screened amplified 43 polymorphic bands.POPGENE analysis showed that Cyathula had a higher level of genetic variation with He of 0.189 2,PPL of 67.4%,Ho of 0.322 4 on the species level,and with He of 0.717,PPL of 49.76%,Ho of 0.250 9 on a population level.Changes in population genetic distances were in the range of 0.52-1.00.CONCLUSION ISSR results reveals that there exist high degree in the species and low degree among population differentiation of Cyathula officinalis,and the genetic variation is mainly caused by cross-pollination.
