摘 要:目的观察吡格列酮对胰岛素抵抗骨骼肌细胞第10号染色体缺失及与张力蛋白同源的磷酸酶(PTEN)及其磷酸化、蛋白激酶B(Akt)磷酸化蛋白表达、细胞凋亡的影响。方法传代培养大鼠骨骼肌细胞,棕榈酸作用制备胰岛素抵抗模型。分以下3组:正常细胞组、胰岛素抵抗组、胰岛素抵抗+吡格列酮组。3组细胞各经胰岛素刺激15 min。Western blotting检测各组PTEN、PTEN磷酸化及Akt磷酸化的表达。流式细胞仪检测各组细胞凋亡率。结果胰岛素抵抗组大鼠骨骼肌细胞PTEN及PTEN磷酸化蛋白表达较正常细胞组增加,差异有统计学意义(P<0.01);胰岛素抵抗组Akt磷酸化蛋白表达较正常细胞组下降,差异有统计学意义(P<0.01);胰岛素抵抗+吡格列酮组PTEN、PTEN磷酸化及Akt磷酸化蛋白表达较胰岛素抵抗组均明显增加,差异有统计学意义(P<0.01)。胰岛素抵抗组与正常细胞组比较凋亡细胞明显增多,差异有统计学意义(P<0.01);胰岛素抵抗+吡格列酮组与胰岛素抵抗组比较凋亡细胞明显增多,差异有统计学意义(P<0.01)。结论胰岛素抵抗时PTEN及其磷酸化表达增加,影响细胞凋亡。吡格列酮通过增加PTEN基因的表达,进一步增加骨骼肌细胞凋亡。【Objective】 To observe the influence of Pioglitazone to insulin resistance skeletal muscle cells on phosphatase and tensin homolog deleted on Chromosome ten(PTEN),the protein kinase B(Akt) phosphorylated proteins expression and cell apoptosis.【Methods】 Subculturing the skeletal muscle cells of rats,and the hexadecanoic acid act on them to prepare insulin resistance model.Divided into three groups: common cells group,insulin resistance group and insulin resistance + Pioglitazone group.Three groups of cells were being stimulated by insulin for 15 min.PTEN,p-PTEN,p-Akt expression of each group were tested by Western blotting.The apoptosis rates were teste by flow cytometry.【Results】 The PTEN and p-PTEN protein expressions of insulin resistance group compared with the normal cell group were increased,the difference was statistically significant(P〈0.01).The p-Akt protein expression of insulin resistance group compared with the normal cell group was decreased,the difference was statistically significant(P〈0.01).The PTEN,p-PTEN andp-Akt protein expression of insulin resistance + Pioglitazone group compared with the insulin resistance group were all increased,the differences were statistically significant(P〈0.01).The number of apoptotic cells of insulin resistance group compared with the normal cell group was increased,the difference was statistically significant(P〈0.01).The number of apoptotic cells of insulin resistance + Pioglitazone group compared with the insulin resistance group was increased,the difference was statistically significant(P〈0.01).【Conclusion】 When insulin resistance,PTEN and its phosphorylation expressions were increased and it effected apoptosis.The Pioglitazone increased apoptosis in skeletal muscle cells by increasing the expression of PTEN.
