摘 要:目的建立不同断面颜色土茯苓药材的薄层色谱指纹图谱。方法高效硅胶G预制板点样1μL,展开剂为甲苯-乙酸乙酯-甲酸(6∶5∶1.5),饱和20 min,展距8 cm,显色喷以5%三氯化铝乙醇溶液,放置5 min;扫描波长320 nm;狭缝6.0 mm×0.3 mm,速度20 mm.s-1。结果红棕色断面样品具有6个共有峰,相似度>0.95;类白色断面样品具有4个共有峰,相似度>0.90;在薄层图谱上二者具有显著性差异。结论所用方法简单且易于操作,专属性强,可用于区分不同断面颜色的土茯苓药材,为区分红棕色断面和类白色断面的土茯苓提供了科学依据。OBJECTIVE To establish thin layer chromatography(TLC) fingerprint of medical materials of Smilacis rhizoma with different section colors.METHODS 1 μL spotting solution was put on the precast slab of silica gel G with high performance.benzene-ethyl acetate-formic acid(6:5:1.5) was used as the developer.Saturation time was 20 min and the developed height was 8 cm.Color reaction happened in 5 minutes when 5% solution of aluminium chloride and ethanol was sprayed on the precast slab.The scanning wave-lengths was 320 nm and slit width was 6.0 mm×0.3 mm,and the speed was 20 mm·s-1.RESULTS The sample of reddish brown section had 6 common peaks and the similarity degree was more than 0.95.The samples of nearly white section had 4 common peaks and the similarity degree was more than 0.90.There are significant difference of them in TLC maps.CONCLUSION This method is simple and easily operated with good specificity and can be used for distinguishing different medical materials of Smilacis rhizoma with different section colors.It provides scientific basis for distinguishing medical materials of Smilacis rhizoma with different section colors including reddish brown and nearly white.
