白芷提取物的HPLC指纹图谱研究

摘　　要：目的建立白芷提取物HPLC指纹图谱分析方法。方法采用Diamonil C18色谱柱(250 mm×4.6 mm,5μm),流动相为甲醇-水-四氢呋喃,梯度洗脱,流速1.0 mL.min-1,检测波长320 nm。结果 10批未熏硫白芷制备的提取物HPLC指纹图谱相似度均在0.9以上,确定了9个峰作为特征峰组成其对照指纹图谱,并对其中的8个峰进行了指认,分别为异紫花前胡内酯、8-甲氧基补骨脂素、佛手内酯、白当归脑、氧化前胡素、欧前胡素、水合氧化前胡内酯、异欧前胡素。熏硫白芷制备的提取物与对照HPLC指纹图谱比较,有色谱峰的大幅度降低或缺失。结论建立的指纹图谱较全面地反映了白芷提取物中香豆素类成分的特征,分析方法简便、重复性好,为有效地控制白芷提取物的质量提供了新的评价方法和依据。OBJECTIVE To establish an HPLC fingerprint analysis method for Angelicae dahuricae extracts.METHODS HPLC fingerprint was performed on Diamonil C18 column（250 mm×4.6 mm,5 μm） with mixture of methanol-water-THF as mobil phase in a gradient mode at a flow rate of 1.0 mL·min-1.The detection wavelength was 320 nm.RESULTS 10 batch of extracts prepared by non-sulfur-fumigated Angelica dahurica HPLC fingerprint had 9 characteristic common peaks and their similarity was above 0.9.8 peaks were identified,they were marmesin,xanthotoxin,bergapten,byakangelicol,oxypeucedanin,imperatorin,oxypeucedanin hydrate and isoimperatorin.Peaks from extracts prepared by sulfur-fumigated Angelica dahurica HPLC fingerprint are greatly reduced or absent.CONCLUSION The fingerprints can comprehensively reflect the characteristics of coumarins in Angelicae dahuricae extracts.The method is convenient,reproducible which provided a new evaluation method and basis for the effective control of the quality of Angelicae dahuricae extracts.
