Property of Thermosensitive PMMA Encapsulated Hollow Fe_3O_4 Magnetite Nanoparticles for Drug Delivery

摘  要:利用聚甲基丙烯酸甲酯(PMMA)包覆空心Fe3O4磁粒制备了温敏性Fe3O4@PMMA复合粒子,并采用对氨基水杨酸(Mr=153.14)作为模拟负载药物,考察了不同温度与pH对复合粒子的接枝、包覆、载药与控释的影响。载药与控释研究结果表明,磁粒空心结构及PMMA膜与药物之间的氢键作用与温敏效应,显著提高了药物负载量,每0.0100g复合磁粒可负载1468μg(0.959×10-5mol)药物。同时,超过80%的药物在3h^4h内释放,具有局部高浓度药物释放性能,并且药物最大释放量与时间呈线性关系。这种复合空心磁粒载药体系的构建有效提高了药物负载效率,配合磁靶向性能,将在癌症载药治疗方面展现巨大的应用前景。Thermosensitive polymethylmethacrylate(PMMA) encapsulated hollow Fe3O4 nanoparticles Fe3O4@PMMA composites were fabricated.p-Aminosalicylic acid(Mr=153.14) used as the standard drug,the properties of Fe3O4@PMMA composites for drug delivery under the conditions of variant temperature and pH were investigated.The results show that the drug loading capacity is significantly enhanced by the magnetic cavity structure,hydrogen-bonding interaction and temperature-responsive effect between thermosensitive polymer and the drug.The drug loading ratio is about 1468.4 μg(0.959×10-5 mol) standard drug per 0.0100g nanocomposite particles.Meanwhile,about 80% drugs are released within 3 to 4 hours,and the maximized drug delivery content shows the linear correlation with the release time.These findings prove that the hollow magnetic nanostructures and smart thermosensitive polymer encapsulation would be useful for drug delivery system in anticancer biomedical applications.
