摘  要:目的采用多指标正交实验考察黄芪有效部位黄芪黄酮、黄芪多糖、黄芪皂苷对糖尿病模型大鼠的药效机制,分析它们之间的交互关系。方法用正交设计安排实验,将126只大鼠随机分为9组,每组14只。除正常组外,其余各组按52 mg/kg剂量对大鼠进行一次性腹腔注射链脲佐菌素。造模后正常组、模型组给予蒸馏水6.3 mL/kg,其余各给药组分别按照0.105、0.284、0.389、0.043、0.148、0.327、0.432 g/kg剂量喂饲黄芪有效部位药物。造模后30 d测定血糖、血脂、脂联素、白细胞介素等药效、机制、指标。有关联的多项指标计算综合评分,对综合指标进行正交设计方差分析。结果模型组与正常组比较,除低密度脂蛋白外其他各项指标差异均有统计学意义(P<0.05)。黄酮有降血糖作用,多糖、皂苷作用不明显;黄酮、皂苷有降血脂作用,联合用药效果不如单一用药;黄酮、皂苷对脂联素有提升作用,二者合用效果优于单一用药。黄酮、多糖、皂苷对血清白介素有调节作用,黄酮和多糖、皂苷合用效果均优于单一用药。结论黄酮降血糖,黄酮、皂苷降血脂作用明显;在调节脂联素、白细胞介素方面,黄酮与皂苷具有协同作用。Objective To investigate the effective mechanism of effective fractions of Huangqi (Radix Astragali seu Hedysari)-astraisoflavan, astragalus polysaccharide and astragaloside by using multiple indicator orthogonal experiment in rat model of diabetes, and analyze the interactive relationship among them. Methods Arranging experiment with orthogonal design, 126 rats were randomly divided into 9 groups (each n = 14 ). Except normal group, other groups were given intraperitoneal injection of streptozocin (52 mg/kg). After modeling normal and model groups were given distilled water (6.3 mL/ kg) and other groups were given effective fractions of Huangqi (0. 105 g/kg, 0.284 g/kg, 0.389 g/kg, 0.043 g/kg, 0. 148 g/kg, 0. 327 g/kg and 0.432 g ~ kg). The changes of plasma glucose, blood fat, adiponectin (ADPN) and interleukin (IL) were detected 30 days after modeling. The synthetical scoring of relevant multiple indicators was calculated and the multiple indicators were analyzed by using orthogonal design and variance analysis. Results Compared with normal group, except of low-density lipoprotein ( LDL), the difference in other indicators had statistical significance ( P 〈 0.05 ) in modelgroup. Astraisoflavan had effect of decreasing astragaloside had not. Astraisoflavan and astragal plasma glucose, but oside had the effect of astragalus polysaccharide and decreasing blood lipid and the effect of their combination was lower. Astraisoflavan and astragaloside had the effect of increasing adiponectin and the effect of their combination was higher. Astraisoflavan, astragalus polysaccharide and astragaloside had effect of regulating IL and the combination had higher effect. Conclusion Astraisoflavan has significant effect of decreasing plasma glucose, and astraisoflavan and astragaloside have significant effect of decreasing blood fat. In the regulation of ADPN and IL, astraisoflavan and astragaloside have a synthetic effect.
