摘  要:目的观察抗心衰颗粒对舒张性心力衰竭(DHF)大鼠模型心肌细胞内钙离子(Ca2+)、ATP酶及心肌线粒体超微结构的影响。方法用腹主动脉缩窄法建立舒张性心衰大鼠模型,随机分为模型组、阳性药组及抗心衰颗粒大、中、小剂量组,另设假手术组。激光共聚焦检测心肌细胞内Ca2+荧光强度;分光光度法检测Ca2+-ATP酶与Na+-K+-ATP酶活性;电镜检测心肌线粒体超微结构的变化。结果与假手术组比较,模型组的大鼠心肌细胞内Ca2+荧光强度上升,Ca2+-ATP酶及Na+-K+-ATP酶活力下降(P<0.05);与模型组相比,阳性药组及抗心衰颗粒3组细胞内Ca2+浓度和Ca2+-ATP酶活力均有改善(P<0.01);抗心衰颗粒中、大剂量组Na+-K+-ATP酶活力较模型组提高(P<0.01)。电镜下,假手术组心肌线粒体结构清晰,模型组可见明显空泡化,嵴熔合模糊不清,或断裂、缺失,肌浆网终池扩张,阳性药组和抗心衰颗粒治疗组心肌线粒体轻度水肿,病变较模型组轻。结论抗心衰颗粒能提高心肌细胞Na+-K+-ATP酶和Ca2+-ATP酶活力,降低胞内Ca2+浓度,并减轻心肌线粒体病变。提示抗心衰颗粒对DHF大鼠有治疗作用,其机制与心肌细胞钙离子转运和能量代谢相关。Objective To observe the influences of Kangxinshuai Granules on myocardial intracellular calcium ion (Ca2+ ), ATPase and mitochondrial uhrastructure in rats with diastolic heart failure (DHF). Methods The rat model of DHF was established by using abdominal aortic coarctation, and then the rats were randomly divided into model group, benazepril group, high-dose, mid-dose and low-dose Kangxinshuai Granules groups (high-dose group, mid-dose group and low-dose group ) and sham- operation group. The fluorescence intensity of myocardial intracellular Ca2+ was detected by using laser scanning confocal microscope (LSCM), activities of Ca2+ -ATPase and Na2+-K+-ATPase were detected by using spectrophotometry, and c using electromicroscope. Results hanges of myocardial mitochondrial ultrastructure were detected by Compared with sham-operation group, the fluorescence intensity of myocardial intracellular Ca2+ increased and activities of Ca2+ -ATPase and Na2+-K + -ATPase decreased in model group (P 〈 0.05 ). Compared with model roup, the concentration of Ca2+ and activities of C.a2+ -ATPase increased in benazepril group and three Kangxinshuai Granules groups ( P 〈 0. 01 ). The activities of Na2+-K+ -ATPase increased in high-dose group and mid-dose group compared with model group (P 〈 0. 01 ). Under the electromicroscope, mitochondrial ultrastructure was clear in sham- operation group, and showed significant vacuolization and blurred or fractured or deleted crest fusion and dilated sarcoplasmic reticulum terminal cisterna in model group. In benazepril group and three Kangxinshuai Granules groups, myocardial mitochondrial ultrastructure showed mild edema but the pathological changes were less than in model group. Conclusion Kangxinshuai Granules can improve the activities of Na + -K + -ATPase and Ca2 ~ -ATPase, reduce the concentration of myocardial intracellular Ca2+, and decrease the pathological changes indicates that be related to Kangxinshuai Granules had th
