摘  要:目的:研究蕨麻正丁醇部位对缺氧损伤心肌细胞凋亡及Caspase 3、Caspase 9表达的抑制作用。方法:建立大鼠原代培养心肌细胞缺氧模型,HE染色观察细胞组织形态,PI-Hochesst3342染色观察细胞凋亡,RT-PCR技术及免疫细胞化学染色方法检测细胞Caspase 3、Caspase 9 mRNA及其产物蛋白水平。结果:与模型组比较,正丁醇部位各剂量组Caspase 3、Caspase 9 mRAN表达水平均显著降低(P<0.05或P<0.01);高剂量组Caspase 3、Caspase 9蛋白水平显著降低(P<0.01)。结论:蕨麻正丁醇部位可能通过减弱缺氧诱导的心肌细胞Caspase级联反应,抑制心肌细胞凋亡。Objective: To study the inhibting effect of the n- butanol Potentilla anserine L. fraction on the apoptosis and the expressions of Caspase 3 and Caspase 9 of cardiomyocyte injured by hypoxia. Methods: To develop a model of cardiomycyte injured by hypoxia in rats in vitro. The pathology changes and the apoptosis of cardiomycytes were observed separately by HE staining and PI - Hoechst3342. The expression levels of Caspase3 and Caspase9 mRNAs in the cardio- mycytes were studied by reverse transcription polymerase chain reaction( RT - PCR). The localizations and levels of their product proteins in the cardiomycytes were investigated by immunocytochemistry stain. Rersult: Comparing with model group, the pathology changes were improved by the n - butanol Potentilla anserine L. fraction and the number of apoptotic cardiomycytes was reduced. After treated with different doses of n - butanol Potentilla anserine L. fraction, the expres- sion levels of Caspase 3 and Caspase 9 mRANs in cardiomyocytes significantly decreased( P 〈0. 05 or P 〈0. 01 ). The contents of their product proteins significantly decresed in the high dose group( P 〈0. 01 ). Conclusion: The n- butanol Potentilla anserine L. fraction exerts a remarkably inhibiting effect on cardiomyocyte during hypoxia and this may be relat- ed with its effect of attenuating Caspase cascade reaction and inhibiting the apoptosis.
