角花胡颓子提取物对荷瘤鼠肿瘤细胞凋亡的影响

摘　　要：目的:探讨角花胡颓子提取物(JHT)的抑瘤机制,为其开发应用提供实验依据。方法:计算抑瘤率,电镜观察肿瘤细胞超微结构。原位杂交法检测Survivin mRNA表达。结果:GFW抑瘤率为39.02%,透射电镜观察到JHT作用下的肿瘤细胞凋亡的形态学改变,可见凋亡细胞及典型凋亡小体形成。JHT抑制肿瘤细胞Survivin表达,与模型组比较,差异有统计学意义。结论:角花胡颓子提取物具有抑瘤作用,可诱导肿瘤细胞凋亡,其机制可能与下调Survivin mRNA表达相关。Objective ： To study the influence of JHT on tumor apoptosis and to apply the experimenting basis of JHT＇s developing and utilizing. Methods：The S180 mice model was used to detective the inhibiting effects of GFW, observing the ukrastructural change by electron microscopy. The mRNA expression of Survivin was evaluated by situ hybridization. Re- sults：For S180 sarcoma, the inhibiting ratio of JHT was 39.02%. Some typical apoptotic cells and apoptotic bodies were observed through electron microscope. Survivin mRNA was markedly declined in the JHT group when compared with the model group （P 〈 0. 01 ） . Conclusion ：JHT induces the tumor cell＇ apoptosis, its mechanism may be closely related with adjusting expression levels of Survivin gene.
