积雪甙对伤口愈合作用的基础研究

摘　　要：目的:探讨积雪甙治疗伤口愈合的影响。方法:采用MTT法测量积雪甙对角质形成细胞和血管内皮细胞株增殖作用的影响,采用RT-PCR方法测量积雪甙对血管内皮细胞分泌KGF- 2mRNA量的影响。结果:积雪甙在浓度为1.5μg/ml左右时对角质形成细胞和血管内皮细胞促增殖作用最大,在此浓度下血管内皮细胞KGF-2mRNA表达量最大。结论:积雪甙在浓度为1.5μg/ml对伤口愈合促进作用最大。Objective: To research the effect of the asiaticoside to wound healing. Method: MTT method is used to evaluate the asiaticoside to the proliferation of HaCaT cell (human normal immortal keratinocyte line) and EVC - 304 cell (human umbilical vein endotheliocyte line) and RT - PCR method is used to detect the expression of the KGF - 2 mRNA level of endothelioeyte after the stimulation of asiaticoside. Result; When the dose reaches to 1. 5μg/ml or so, the asiaticoside has the greatest ability to increase the HaCaT and ECV-304 proliferation and, with the same dose, the KGF -2mRNA expression of the endotheliocyte is the highest. Conclusion: Asiaticoside can stimulate the proliferation of the keratinocyte and endotheliocyte, it can also increase the KGF - 2 mRNA expression. It is suggested that asiaticoside can speed up the wound healing.
