摘  要:目的 :一步法分析地高辛相对生物利用度和药物动力学参数。方法 :用NONMEM程序包及其新的POSTHOC子模块功能 ,估算地高辛相对生物利用度和药动学参数 ,并与经典的 3P87方法比较。结果 :两者的相对生物利用度一致 ;青年健康受试者口服地高辛的主要药动学参数典型值K10 (h-1)、Vc/F(L)和 β(h-1)分别为 0 .115 ,12 8和 0 .0 18,而老年病人的典型值K10 (h-1)、Vc/F(L)和 β(h-1)分别为 0 .0 40 3,76 .0和 0 .0 148。结论 :老年病人的主要药动学参数明显低于青年健康受试者 ,差异存在显著性。用NONMEM法在取样点数较少时可获得与经典法一致的个体药动学参数。OBJECTIVE:To evaluate relative bioavailability and pharmacokintic parameters of digoxin by one step method.METHODS:To predict the relative bioavailability and pharmacokinetic parameters of digoxin with NONMEM program and its submodule,POSTHOC.RESULTS:There was no statistical significent difference between NONMEM program and 3P87 approach on evaluating relative bioavailability and pharmacokinetics of digoxin in eight Chinese healthy male volunteers.The typical value of mainly population pharmacokinetics, K 10 (h -1 ), V c/ F(L) and β(h -1 ),of digoxin in young healthy volunteers were 0.115 ,128 and 0.018 respectively.The typical value of K 10 (h -1 ), V c/ F(L) and β(h -1 ),of digoxin in aged patients were 0.040 3 , 76.0 and 0.014 8 respectively.CONCLUSIONS:The elimination rate of digoxin in aged patients was significantly lower than the in young healty volunteers.The individual pharmacokinetic parameters of aged patients fitted by NONMEM using POSTHOC module for aparse data( n =3~4) were similar to it obtained by PKBP N1 program for normal data( n =5~7).
