摘 要:目的探讨顺铂(DDP)诱导消化系统迟发性反应机制及中药丹参(SM)的拮抗作用。方法 30只豚鼠随机分为正常对照组、DDP组、丹参+顺铂(SM+DDP)组,观察用药前后体质量变化、胃电图(EGG)改变,并进行胃、肠组织超微结构观察。结果 DDP组豚鼠体质量明显减轻(P<0.05),透射电镜发现胃肠组织细胞破坏严重,而SM+DDP组豚鼠各方面情况要好于DDP组,同时透射电镜表现有大量吞噬体存在。各组胃电图显示DDP组振幅增加、频率增快(P<0.05),而SM+DDP组EGG与正常对照组比较无明显变化。结论顺铂诱导胃肠道迟发性反应的发生可能与胃肠组织黏膜屏障功能受损,引起致病菌侵害有关,而丹参能够清除致病菌,从而具有一定的拮抗作用。Objective To explore the antagonistic mechanism of cisplatin(DDP) on toxicity damage of gas-trointestinal tissue induced by DDP. Methods Thirty healthy guinea pigs were divided into three groups at ran-dom: the control group, DDP group,SM+DDP group. The weight and EGG change of guinea pigs were exam-ined. Ultrastructure change was observed by transmission electron microscope(TEM). Results The body weight of DDP group decreased obviously(P〈:0.05). The EGG amplitude of DDP group increased and the frequency was fast(P〈0. 05) after administration, and the change was serious under the TEM observing; while salvia miltiorrhi- za could prevent the change of EGG caused by DDP, showing the amplitude decreased, and the frequency was slow, and the ultrastructure change was not obvious, and meanwhile phagosome was observed. Conclusion There may be relationship between gastrointestinal tract tardive reaction induced by DDP and stomach intestine mucosa barrier damage, and dysfunction afterwards, while salvia miltiorrhiza could clear pathogenic bacteria and have an-tagonism.
