致病性灿烂弧菌的分离鉴定及药敏特性研究

摘  要:[目的]分离鉴定患病刺参(Apostichopus japonicus)的致病茵,并研究其药敏特性。[方法]从患病的刺参体内分离出1株致病菌W-1。通过微量生化鉴定管和菌体常规形态特征、生理生化反应指标测定以及16S rRNA测序分析等进行综合鉴定;再通过人工感染健康刺参测定该菌株对其的半数致死量(LD50);最后进行药敏试验确定其药敏特性。[结果]致病菌W-1为革兰氏阴性菌,菌落呈米色,边缘光滑,中间略有隆起。序列分析结果显示,该菌株与灿烂弧菌的亲缘关系最近,相似率达99.8%,综合鉴定该菌为灿烂弧菌(Vibrio splendidus)。该菌对苯唑青霉素、头孢氨苄、头孢唑林、头孢拉定、头孢呋新、菌必治、先锋必素、麦迪霉素、氟哌酸、环丙沙星、万古霉素、痢特灵、丁胺卡那、庆大霉素、强力霉素、氯洁霉素等抗生素高度敏感,对氨苄青霉素、羧苄青霉素、氧哌嗪青霉素、复达欣、新霉素、四环素等药物中度敏感。[结论]该研究对防治由灿烂弧菌引起的海水养殖动物疾病具有积极意义。[Objective] The r+esearch aimed to isolate japonicus, and its antibiotic sensitivities were studied and identify the pathogenic bacterial strain from ill Apostichopus [Method] One pathogenic bacterial strain W-1 to sea cucumber, Apostichopus japonicus was isolated. The trace biochemical identification tube, traditional physiological and biochemical experiments and 16S rRNA gene homology comparison of the bacteria were used to identify it. The infecting test of sea cucumbers was used to determine LDso. Then antibiotic sensitivities of the pathogenic bacterial strain were studied. [Result] The result showed that strain W-1 was gram negative, had the similar characters as Vibrio splendidus, having short rods with a single polar flagellum, forming yellow colony on TCBS agar. Further results from 16S rRNA sequence analysis showed that the sequence of bacterium W-1 was highly identical to Vibrio splendidus, with 99.8% identity. Thus bacterium W-1 was identified as Vibrio splendidus. The LD50 of the strain W-1 to sea cucumbers was 10^9 CFU/mL. In addition, strain W-1 was highly sensitive to oxacillin, cephalothin IV, cephalothin V, cephalothin VI, ceftriaxone, cefoperazone, midecamycin, norfloracin, ciprofloracin, vancomycin, furazolidonum, amikacin, gentamycin, doxycicline hyclate, clindamycin and intermediately sensitive to six kinds of antibiotics including amoxicillin, carbenicillin, piperacillin, fortum, neomycin, tetracycline. [Conclusion] The study has positive meaning to defend and control sea cucumber diseases caused by Vibrio splendidus.
