摘 要:目的通过观察白藜芦醇对甲状腺鳞癌细胞株SW579生长及CDC25B蛋白(cell division cycle 25B)表达的影响,探讨白藜芦醇的抗癌作用机制。方法取对数生长期SW579细胞培养12h后,按浓度梯度25、50、100、200、400μmol.L-1加入白藜芦醇(实验组),对照组加入等体积含对应浓度DMSO的培养基,应用噻唑蓝(MTT)比色法和流式细胞术检测细胞增殖和细胞周期的变化,同时应用Western Blot方法检测CDC25B蛋白表达情况。结果不同浓度(25、50、100、200、400μmol.L-1)白藜芦醇对SW579细胞生长均有抑制作用,且呈剂量依赖性(P<0.05),并且改变细胞周期分布,实验组(100μmol.L-1白藜芦醇)S期[(28.55±0.58)%]比例明显高于对照组[(19.49±1.27)%](P<0.05),G2/M期[(8.79±0.74)%]比例显著低于对照组[(15.88±0.81)%](P<0.01),细胞分裂被阻滞。白藜芦醇作用24h后,与对照组(0.421±0.004)相比,实验组(100μmol.L-1白藜芦醇)(0.062±0.012)CDC25B蛋白表达明显下降(P<0.01)。结论白藜芦醇能够显著抑制SW579细胞株的增殖,使其处于S期阻滞,其抑制作用可能与CDC25B蛋白表达下降有关。Objective To observe the effects of resveratrol on cell proliferation and CDC25B protein expression in thyroid squamous cell carcinoma SW-579 cells,and to investigate the mechanism of its action.Methods After culture for 12 hours,SW-579 cells in the logarithmic growth phase were treated with different concentrations of resveratrol(25,50,100,200 and 400 μmol·L-1).Control cells were given an equivalent volume of DMSO.Cell proliferation and cell cycle were measured by MTT assay and flow cytometry,respectively.The expression of CDC25B protein was detected by Western blot.Results The treatment with resveratrol significantly inhibited the proliferation of SW-579 cells in a dose-dependent manner(P〈0.05).Furthermore,resveratrol treatment altered cell cycle distribution and induced cell cycle arrest.Compared with the control,resveratrol(100 μmol·L-1) obviously increased the percentage of cells in S phase [(28.55±0.58)% vs(19.49±1.27)%](P〈0.05),but markedly decreased the percentage of cells in G2/M phase [(8.79±0.74)% vs(15.88±0.81)%](P〈0.01).In addition,resveratrol(100 μmol·L-1) significantly reduced the expression of CDC25B protein compared with the control(0.062±0.012 vs 0.421±0.004,P〈0.01).Conclusion Resveratrol can inhibit cell proliferation and induce S phase arrest in SW-579 cells through down-regulating the expression of CDC25B protein.
