摘　　要：目的了解饲喂Ⅱ型胶原(typeⅡcollagen ,CⅡ)的大鼠能否在其肠相关淋巴组织和脾脏中 ,诱导产生CⅡ特异性T细胞及其分泌的细胞因子的类型。方法对口服CⅡ致耐大鼠进行特异和非特异性淋巴细胞增殖实验 ,并采用酶联免疫斑点法 (ELISPOT) ,对CⅡ特异性T细胞在单细胞水平进行细胞内细胞因子检测。结果CⅡ致耐大鼠的T细胞增殖能力下降 ,CⅡ特异性T细胞多数分泌TGF β,极少数分泌IL 4和IFN γ。结论多次口服小剂量CⅡ可在佐剂性关节炎(adjuvantarthritis ,AA)大鼠模型中 ,诱导产生分泌TGF β的CⅡ特异性T细胞(Th3细胞) ,抑制AA的进展。Aim To understand whether or not type Ⅱcollagen (CⅡ) specific T cells can be induced by oral administration of CⅡand which kinds of cytokines are secreted by the cells. Methods Specific and non specific lymphocyte proliferation test and enzyme linked immunospot(ELISPOT) assay were used in the present study. Results An increase in the CII reactive TGF β secreting T cell lines occurred in CII treated rats as compared with un treated rat. Conclusion Oral administration of low doses of CII could suppress the development of adjuvant arthritis(AA) and downregulate Th1 cell mediated systemic DTH by secreting the Th1 suppressive cytokine, such as TGF β.
