摘 要:目的探讨复方薤白胶囊对野百合碱所诱导的肺动脉高压大鼠模型血管细胞黏附因子(VCAM-1)和细胞间黏附因子(ICAM-1)的影响。方法 SD大鼠随机分为正常对照组、模型组、复方薤白胶囊高、低剂量组。腹腔注射野百合碱制作肺动脉高压动物模型,于造模第2日开始灌胃给药,给药7周后测定其平均肺动脉压(mPAP)和平均右心室压(mRVP),采用免疫组化法测定肺组织VCAM-1和ICAM-1的表达。结果 (1)模型组的mPAP和mRVP较正常对照组明显升高(P<0.01),复方薤白胶囊高剂量组和低剂量组mPAP和mRVP较正常组高但较模型组则显著下降(P<0.05);(2)与正常对照组相比,其余各组的VCAM-1和ICAM-1含量明显增加(P<0.05);与模型组相比,复方薤白胶囊高剂量组的VCAM-1和ICAM-1含量显著下降(P<0.05),低剂量组VCAM-1和ICAM-1含量与模型组相比则无显著差异。结论 VCAM-1和ICAM-1的增加与肺动脉高压的形成相关;复方薤白胶囊能有效降低野百合碱诱导肺动脉高压大鼠的肺动脉压可能与其减少VCAM-1和ICAM-1的表达有关。Objective: To explore the effect of compound Macro-stem Onion capsule on the expression of vascular cell adhesion molecule-1(VCAM-1) and intercellular cell adhesion molecule-1(ICAM-1) in the lung of rat models with pulmonary hypertension.Methods: SD rats were randomly divided into four groups including the normal control group,the model group,the high dose of compound Macro-stem Onion capsule group and the low dose of compound Macro-stem Onion group.Animal model of pulmonary hypertension was made by intra-peritoneal injection of wild lily alkali(MCT).2 days later,drug were given by intra-gastric injection.Average pulmonary arterial pressure(mPAP) and average right ventricular pressure and average(mRVP) were determined after 7 weeks.The expressions of VCAM-1 and ICAM-1 in the lung tissues were determined by immunohistochemical method.Results: Compared with the normal control group,the mPAP and mRVP of model group were obviously higher(P 0.01).The mPAP and the mRVP of high dose and low dose of compound Macro-stem Onion capsule groups were significantly higher than those of the normal control group and the lower than the model group.Compared with the normal control group,the expressions such as the VCAM-1 and the ICAM-1 of other groups were increased significantly.Compared with the model group,expressions including VCAM-1 and ICAM-1 of high dose of compound Macro-stem Onion capsule group were obviously decreased.The difference between the model group and the low dose of compound Macro-stem Onion group was not significant.Conclusion: The increases of VCAM-1 and ICAM-1 may be related with the formation of pulmonary arterial hypertension.Meanwhile,compound Macro-stem Onion capsule can effectively reduce pulmonary arterial pressure which may be associated with reduced expression of VCAM-1 and ICAM-1.
