摘  要:目的研究丹参素配伍川芎嗪对大鼠局灶性脑缺血的保护作用。方法采用线栓法制备SD大鼠永久性大脑中动脉阻塞(permanent middle cerebral artery occlusion,pMCAO)模型。实验SD大鼠随机分为6组,分别为假手术组、模型组、丹参素组(1.2 mg·kg-1)、川芎嗪组(60mg·kg-1)、丹参川芎嗪注射液组[(1.2mg丹参素+60mg川芎嗪)kg-1]、尼莫地平注射液组(1.2mg·kg-1)。造模24 h后,测定神经行为学评分、脑组织含水量、脑梗塞体积、血清中超氧化物歧化酶(SOD)活性和兴奋性氨基酸谷氨酸(Glu)含量,并进行组织病理学检查。结果与模型组相比,丹参素组、川芎嗪组、丹参川芎嗪注射液组大鼠的神经行为明显改善,脑含水量和脑梗塞体积显著降低(P<0.05或0.01),血清中SOD活性升高,Glu含量降低(P<0.05或0.01),HE染色显示组织间隙较小,病理损伤小于模型组。丹参川芎嗪注射液组保护作用最优,说明两单味药配伍使保护作用增强。结论丹参素配伍川芎嗪对大鼠局灶性脑缺血有协同的保护作用。Objective To investigate the protective effect of tanshinol(TSN) combined with tetramethylpyrazine(TMP) on focal cerebral ischemia in rats.Methods Permanent middle cerebral artery occlusion(pMCAO) rat model was established with suture method.The rats were randomly divided into 6 groups: a sham-operated group,a model group,a TSN group(1.2mg·kg-1),a TMP group(60mg·kg-1),a TSN + TMP group [(1.2mg TSN + 60mg TMP) kg-1],and a nimodipine group(1.2mg·kg-1).At 24h after the surgery,the neurological behavior score,brain water content,and infarct volume were measured.Superoxide dismutase(SOD) activity and glutamate(Glu) content in the serum were measured.Histopathological examination was conducted.Results Compared with the model group,the TSN,TMP and TSN + TMP groups alleviated the neurological behaviors,decreased the brain water content and infarct volume(P 0.05,P 0.01),increased the serum SOD activity and lowered the serum Glu content(P 0.05,P 0.01).HE staining showed that the tissue space was narrowed and edema reduced.The effect of TSN + TMP group was better than single medicine,showing certain synergic effect.Conclusion The combination of tanshinol and tetramethylpyrazine synergicall protects the focal cerebral ischemia in rats.
