摘 要:为探讨皖南花猪卵巢组织脂联素受体mRNA与雌二醇(E2)合成相关因子mRNA表达的发育性变化及其相关性,用ELISA法测定0、30、45、90和180日龄(n=5)皖南花猪雌性仔猪血液脂联素(Adp)和E2水平,荧光实时定量PCR法检测卵巢组织脂联素受体1、2(adiponectin receptor 1,2;AdpR1,AdpR2)和卵泡刺激素受体(FSHR)、芳香化酶P450(aromatase P450arom)基因CYP19 mRNA的表达水平。试验结果显示:雌性仔猪血液E2质量浓度、卵巢组织AdpR1、AdpR2和FSHR mRNA表达均有显著或极显著的发育性变化(P<0.05或P<0.01)。血液Adp质量浓度在30 d较低,90 d较高;血液E2质量浓度在0 d最高,30 d较低。AdpR1和AdpR2 mRNA表达量均表现为30和90 d较高,0、45和180 d较低;FSHR mRNA表达量在45 d达到最高,90 d降至最低;CYP19 mRNA表达量在0至45 d较低,90 d时最高。卵巢组织中AdpR1 mRNA表达与AdpR2mRNA表达显著正相关(r=0.380,P<0.05);AdpR1 mRNA表达与CYP19 mRNA表达显著负相关(r=-0.472,P<0.05),与血清E2水平显著负相关(r=-0.416,P<0.05)。结论:Adp可能参与对E2合成(或分泌)的调节。Wannanhua gilts were used in order to observe the developmental patterns and the correlation of adiponectin receptors and estradiol synthesis-related factors mRNA expressions in ovary.Five Wannanhua gilts were sampled at 0,30,45,90,and 180 days of age respectively.The concentrations of serum Adp and serum E2 were measured by ELISA.The expressions of AdpR1,AdpR2,FSHR,and CYP19 mRNA in ovary were detected by real-time fluorescence quantitative RT-PCR.The results showed that the serum Adp and serum E2 concentration,and the mRNA expression of AdpR1,AdpR2 and FSHR in ovary of Wannanhua gilts changed with age increasing(P<0.05 or P<0.01).The serum Adp level decreased at 30 days of age,and increased at 90 days of age.The concentration of serum E2 was highest at 0 day of age,and decreased at 30 days of age.The expressions of AdpR1 and AdpR2 mRNA were both higher at 30 and 90 days of age and lower at 0,45 and 180 days of age.The expression of FSHR mRNA reached the peak at 45 days of age and decreased at 90 days of age.CYP19 mRNA expressions was lower during 0 to 45 days of age,and increased at 90 days of age.Significant positive correlation was found between mRNA expression of AdpR1 and AdpR2 mRNA expression in ovary(r=0.380,P<0.05).Significant negative correlation were found between AdpR1 mRNA expression and CYP19 mRNA expression(r=-0.472,P<0.05)or between AdpR1 mRNA expression and the concentration of serum E2(r=-0.416,P<0.05)respectively.Conclusion:Adp may participate in regulation of E2 synthesis or secretion.
