摘  要:目的:研究脂联素基因(APM1)启动子区域单核苷酸多态性(single nucleotide polymorphism,SNP)与大理白族2型糖尿病(T2DM)的相关性。方法:运用PCR直接测序技术检测白族正常对照及T2DM患者APM1启动子上的5个SNPs位点:11426A>G、11391G>A、11377C>G、11156insCA、11043C>T的多态性。结果:(1)11391G>A和11043C>T两个位点在白族人群中高度保守;(2)11156insCA位点在正常人群中无碱基插入现象,仅在T2DM患者中存在,但频率非常低;(3)11426A>G和11377C>G位点在两组人群中差异无统计学意义;(4)11426A>G和11377C>G位点相连锁,但在两组人群中差异无统计学意义。结论:APM1启动子上11426A>G、11391G>A、11377C>G、11156insCA、11043C>T这5个多态性位点与大理白族2型糖尿病发生无关。Objective To explore the association of single nucleotide polymorphisms (SNPs) at the proximal promoter region of adiponectin gene (APM1) with type 2 diabetes mellitus (T2DM) in the Bai nationality in Dali. Methods The polymorphisms of APM1 (11426A 〉 G, 11391G 〉 A, 11377C 〉 G, 11156insCA, and 11043C 〉 T) were detected by direct sequencing in nondiabetic controls and patients with T2DM. Results No polymorphisms were found at APMI -11391 and -11043. 11156insCA only existed in T2DM group, with a extremely lower rate of frequency. The frequencies of diversified genotypes and alleles at APM 1 -11426 and -11377 did not differ significantly between the control group and the T2DM group (P 〉 0.05 for all comparisons). Conclusions 11426A 〉 G, 11391G 〉 A, 11377C 〉 G, 11156insCA, and 11043C 〉 T at the proximal promoter region of APM1 is not associated with the pathogenesis of type 2 diabetes.
