不同生长阶段家蚕1-脱氧野尻霉素富集与食桑量的相关性研究

摘　　要：为了研究家蚕富集1-脱氧野尻霉素(DNJ)的机制,采用柱前衍生化高效液相色谱法分析不同生长阶段家蚕及其代谢产物和所食桑叶中DNJ的含量,研究家蚕幼虫体1-脱氧野尻霉素富集与食桑量的相关性。结果显示不同生长期家蚕及其副产物中DNJ的累积量有较大差异,且呈一定规律性变化,先上升后下降,于四龄时最高。家蚕幼虫期蚕体及其副产物中DNJ总累积量与其在该阶段所食桑叶中DNJ总量相当,且家蚕的任一成长发育阶段(蚁蚕和熟蚕不吃桑叶除外),其蚕体DNJ含量均大大高于桑叶中的DNJ含量,表明家蚕具有累积或富集DNJ的机能,本研究结果为进一步研究家蚕富集DNJ的机制提供了理论依据。The objective of this study was to investigate the enrichment mechanism of the 1-deoxynojirimycin （DNJ） in the larvae of the silkworm （Bombyx mori）. Pre-column derivatization coupled with reversed-phase high performance liq- uid chromatography （HPLC） was employed to determine the DNJ content in different growth stage of silkworm larva and its byproducts as well as the content in mulberry leaves. The correlation between the consumption of mulberry leaves and the enrichment of DNJ in the larva of the silkworm was then analyzed. The results showed that the accumulation of DNJ in Bombyx mori and their byproducts in different growth periods were remarkably different, and changed regularly, which was rising first and then to decline, the accumulation reached the peak at the 4th instar. The DNJ content of silkworm body and its＇ byproducts was roughly the same as the content in mulberry leaves that silkworm ate. The DNJ content of silkworm was much higher than the DNJ content of mulberry leaves in different growth stage of silkworm larva. This study revealed that the silkworm had the function of enriching and accumulating DNJ. The results provided the basis for further research of studying the DNJ enrichment and metabolism in the larvae of the silkworm.
