摘 要:为了提高银杏细胞黄酮糖苷的提取得率,以人工培养细胞为对象,利用响应面法(RSM)优化了醇法提取工艺.在单因素基础上,采用Design Expert 8.0软件中的Plackett-Burman设计,筛选出显著因素为:提取时间、提取温度和乙醇体积分数,并拟合得到二次多项式模型.细胞黄酮最佳提取条件为:提取时间3.285h、提取温度76.85℃、乙醇体积分数72.05%.在此提取条件下,利用高效液相色谱法(HPLC)验证总黄酮糖苷提取得率为1.810 1%,与模型预测值一致,说明利用RSM优化提取条件,建立的模型预测银杏细胞黄酮糖苷准确可靠,重复性好.体外抗氧化实验结果表明:黄酮糖苷能较好地清除羟自由基,抑制减缓植物油脂的自氧化过程.For improving the yield of flavone glycosides from cultured Ginkgo biloba cell,the optimization of the ethanol extraction was performed with response surface methodology(response surface methodology,RSM).Results indicate that extract time,temperature and ethanol content have significant effects on extraction rate on the basis of univariate experiments determined by Plackett-Burman design of software Design Expert 8.0.Three factors above can be fitted into a second-order polynomial model.The best combinations achieved are with 72.05% ethanol and water for 3.285 h at 76.85℃.The yields of flavone glycosides experimentally with HPLC is 1.8101%,which is identical with the predictive value.The establishment of the model provides adequate accuracy and repeatability in predicting the yield of flavone glycosides from ginkgo cell.The results of antioxidant experiments in vitro show that the clearance of free radical and inhibition of selt-oxidant of plant oil is ideal.
