摘 要:目的实验研究硫化氢(H2S)对烧伤大鼠肠道组织干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)含量的影响,探讨H2S在烧伤大鼠肠道黏膜屏障功能中发挥的作用。方法将104只健康雄性SD大鼠随机分为烧伤组(96只)和正常对照组(8只),其中烧伤组又分为单纯烧伤组(n=32)、炔丙基甘氨酸(PPG)干预组(n=32)、硫氢化钠(NaHS)干预组(n=32)。各烧伤组分别于伤后2d、7d、14d、21d四个时相点取8只SD大鼠回肠组织,匀浆后取上清液用ELISA方法检测各组大鼠肠道组织中IFN-γ、TNF-α的含量变化情况。正常对照组适应性饲养1周后取回肠组织,检测IFN-γ、TNF-α的含量。结果与正常对照组相比,单纯烧伤组、NaHS干预组、PPG干预组各时相点TNF-α和IFN-γ水平均明显升高(P<0.05),在烧伤后2d TNF-α水平最高,烧伤后14d IFN-γ水平达到最高值,之后呈逐渐下降趋势。NaHS干预组各时相点TNF-α水平较单纯烧伤组均明显降低(P<0.05),而IFN-γ水平明显升高;PPG干预组各时相点TNF-α水平较单纯烧伤组均升高,差异有统计学意义(P<0.05),而IFN-γ均降低。结论 H2S可以显著降低烧伤大鼠肠道组织TNF-α水平,升高IFN-γ的水平,表明H2S可减轻肠道炎性反应,保护肠道黏膜屏障作用,为烧伤后应用外源性H2S加强肠道黏膜屏障功能提供了新思路。Objective To investigate effects of hydrogen sulfide (H2S ) on the activities of interferon-gamma (IFN-γ) and tumor necrosis factor-alpha (TNF-α) in the intestinal tissue of burned rats, in order to explore the function and mechanism of H2S in the intestinal mucosal barrier of burned rats.Methods One hundred and four male SD mices were randomly divided into simply burned group (32 cases) ,sodium hydrosulfide (NaHS,H2S donor ) intervention group (32 cases ) ,the propargylglycine (PPG,a CSE inhibitor) intervention group (32 cases) and the control group (8 cases) .The illeal of the control group was collected after feeding a week,and at 2 d,7 d,14 d and 21 d after burned,eight rats of other three group were detected TNF-α and IFN-γ concentration from ileal tissue by ELISA.Results Compared with the control group,all the levels of TNF-α and IFN-γ increased significantly on every time of the rest 3 groups (P0.05) .The activity of TNF-α was higher than the others on the 2 post-burn day,the level of IFN-γ was the highest on the 14th day,and decreased gradually thereafter.Compared with the simply burned group,the expressions of TNF-α significantly decreased with NaHS content (P0.05) ,and the level of IFN-γ increased.However,the level of TNF-α increased conspicuously,and the activity of IFN-γ decreased with PPG intervention on every time (P0.05) .Conclusion H2S could significantly decrease the activities of TNF-α,and induce the expression of INF-γ in the intestinal tissue of burn rats.H2S could lessen inflammation and protect the intestinal mucosal barrier,in order to give a new idea for using NaHS to promote the function of intestinal mucosal barrier after burned.
