摘 要:目的 观察一氧化氮 (NO)在实验性骨关节炎 (OA)关节滑液中的动态变化 ,并探讨NO在实验性OA软骨细胞凋亡中的作用。方法 用原位末端标记法以及镀铜镉还原法对实验性OA软骨细胞凋亡及关节滑液中的NO含量进行检测。结果 实验性OA中NO水平显著高于正常对照组 ,且随制动时间延长 ,NO水平逐渐升高 ,6周达高峰。此时也出现较多软骨细胞凋亡现象 ,应用一氧化氮合酶 (NOS)抑制剂L NAME后 ,关节液中NO含量降低 ,软骨细胞凋亡现象减轻。结论 NO是引起实验性OA中软骨细胞凋亡的重要介质之一。Objective To observe the effects of dynamic change of nitric oxide (NO) in synovial fluid (SF) in experimental OA and investigate NO and nitric oxide synthetase (NOS) inhibitor L NAME participates in apoptosis of chondrocytes in experimental OA.Methods Apoptosis of chondrocytes was determined by TUNEL and the levels of NO in SF by reduction.Results The level of NO in experimental OA was higher than that of normal control group.As time of immobilization was prolonged,the levels of NO were gradually increasing,and got the hightest piont at week 6.After L NAME was applied introvenously,the NO centents and the TUNEL positive chondrocytes were decreased.Conclusion The NO produced by chondrytes is involved in the process of cartilage lesions.It is a major trigger for apoptosis of chondrocytes in experimental OA.
