摘 要:目的研究大鼠腓肠肌失神经支配后miRNA-1(miR-1)、miRNA-133(miR-133)的表达规律,并分析miR-1、miR-133之间的关系。方法 108只SD大鼠,随机分成9大组,每大组再分实验组和对照组,实验组大鼠切断右下肢坐骨神经,左下肢行坐骨神经探查术,对照组大鼠双下肢均行坐骨神经探查术,在各个时间点取下腓肠肌,采用RT-PCR检测miR-1和miR-133并描述其表达规律。透射电镜观察肌卫星细胞超微结构变化,采用方差分析和LSD法进行统计学分析。结果随着失神经时间的延长,骨骼肌中miR-1和miR-133表达先逐渐下调再升高,2周表达最低,miR-1约为正常时的0.25倍[(0.25±0.12),t=2.771,P=0.018],miR-133为正常时的0.3倍[(0.30±0.19),t=0.076,P=0.018],随后二者表达逐渐升高。在腓肠肌超微结构中,随着失神经时间的延长,肌丝肌节逐渐紊乱,肌小节长度减少,肌质网扩张,线粒体肿胀,线粒体嵴变短,直至消失。结论 miR-1及miR-133表达改变与失神经骨骼肌萎缩发生的机制密切相关。Objective To investigate the expression patterns of miR-1 and miR-133 in the myocytes of denervated gastrocnemius in rat; and to analyze the relationship between miR-1 and miR-133. Methods 108 SD rats were randomly divided into 9 groups (n=12 for each group), including experimental group and control group. For every experimental group the right sciatic nerve was cut off, the left sciatic nerve received sham operation. In control group both the right and the left sciatic nerve received sham operation. Real-Time PCR was used to detect the miR-1 and miR-133 level for each sample. Transmission Electron Microscope was used to observe the morphological of muscle satellite cell. Analysis of Variance and LED was used for statistical analysis. Result The expression of miR-1 and miR-133 begin to decrease gradually first and increased slowly later. The expression of miR-1 was about 0.25[(0.25 ~ 0.12), t = 2.771, P = 0.018] folds of control after 2 weeks' denervation, and the expression of miR-133 was about 0.3[(0.30 4- 0.19), t = 0.076, P = 0.018] folds of control. Both of them began to increase then. In the ultrastructure of myofibers, myoneme and sarcomere became disorder, the length of sarcomere decreased, sarcoplasmic reticulum expanded, mitochondria swelled and mitochondria crista got shorter or even disappeared. Conclusions The expression pattern of miR-1 and miR-133 was closely related to the process of muscle atrophy after denervation.
