摘  要:目的观察止消通脉宁对自发性2型糖尿病KKAy小鼠肾组织转化生长因子β1(TGF-β1)、结缔组织生长因子(CTGF)和骨形成蛋白7(BMP7)和基因表达的影响。方法将16周龄SPF级KKAy小鼠50只,随机分为模型组、缬沙坦组,止消通脉宁高、中、低剂量组,每组10只,10只16周C57BL/6小鼠作为正常对照组。药物组分别灌胃相应浓度药物,正常组与模型组用生理盐水灌胃,每日1次,连续28天。测量小鼠空腹血糖(FBG),实验结束时取血并记录肾重,待测TGF-β1、CTGF和BMP7和基因水平。结果与正常组比较,模型组FBG、肾重和肾小球硬化指数和肾间质损伤评分均升高,差异有统计学意义(P<0.01),肾组织糖尿病肾病(AGEs)蛋白表达增多,TGF-β1、CTGF蛋白和基因表达显著增多,而BMP7表达减少。结论止消通脉宁抑制体内AGEs蓄积,促进抗纤维化因子BMP7表达,抑制促纤维化TGF-β1、CTGF因子表达,改善晚期糖基化终产物(DN)肾损伤可能是其治疗DN的作用机制之一。Objective To observe the effect of Zhixiaotongmaining(ZXTMN) on transforming growth factor-β1(TGF-β1),connective tissue growth factor(CTGF) and bone morphogenic protein 7(BMP7) and genes expression in renal tissue of type 2 diabetes mellitus KKAy mice.Methods Fifty SPF KKAy mice aged 16 weeks old were randomly divided into a model group,a valsartan group,and a high-,medium-and low-dose of ZXTMN group(HZ,MZ and LZ group),ten mice in each one,while ten C57BL/6 mice aged 16 weeks old were selected into a normal control group.The medicine groups were intragastric administrated with corresponding concentration of medicines while the normal control group and the model group with normal saline,once a day for 28 consecutive days.The fasting blood-glucose(FBG) was measured;the blood was collected and kidney weight was recorded after the experiment for detecting TGF-β1,CTGF and BMP7 and gene level.Results Compared with those in the normal control group,FBG,kidney weight,glomerulosclerosis index and renal interstitium injury score in the model group were all increased,with significant difference(P 0.01),the expressions of advanced glycation end products(AGEs),TGF-β1,CTGF and gene level were increased,while the expression of BMP7 was decreased.Conclusion ZXTMN could restrain the accumulation of AGEs,promote the expression of anti fibrosis factor BMP7 and inhibit the TGF-β1,CTGF.The improvement on kidney injury of AGEs might be one of its mechanisms on treating diabetic nephropathy.
