摘 要:【目的】从分子水平了解山东牛心柿群体的遗传结构和亲缘关系。【方法】采用SSR分子标记技术对山东省内的8个牛心柿群体共207个个体进行检测,应用POPGENE 1.31软件进行遗传参数分析,采用AREQUIN 2.0软件对群体间和群体内的遗传变异进行分子变异分析,利用TFPGA软件对Nei’s无偏差预期杂合度和基因分化系数进行了统计计算。【结果】10对SSR引物共检测到128个位点,其中105个位点为多态位点,占82%。Nei’s遗传多样性指数(He)在0.153 8~0.247 6,Shannon信息指数(H0)在0.221 7~0.372 1。群体间的遗传变异占总遗传变异的24.17%(P〈0.001),群体间的遗传距离(D〈0.231 7)、遗传一致度(I〉0.783 2)、基因分化系数(φST=0.222 6)和遗传分化系数(FST=0.175 9)均表明各群体间产生了一定程度的遗传分化。【结论】山东牛心柿群体遗传背景丰富,群体间一定程度的遗传分化可能是人为选择压力所引起。[Objective] The objective of the experiment is to understand the diversity and genetic structure of oriental persimmon (Diospyros kaki) populations in Shandong. [Method] The genetic diversity of 207 individuals from 8 persimmon (Diospyros kaki) populations in Shandong were investigated using SSR markers. The genetic parameters were analyzed using POPGENE 1.31 software, genetic variation of inter- population and inner-population were analyzed by AREQUIN 2.0 version, and then the mean expected heterozygosity (He) and genetic differentiation coefficient were counted by TFPGA software. [Result] As a result, 128 bands, of which 105 were polymorphic loci, were amplified by 10 SSR primers selected from 51 markers. The relatively high levels of PPL (82%), NeFs gene diversity (Heranged from 0.153 8 to 0.247 6) and Shannon's information diversity (H0 ranged from 0.221 7 to 0.372 1) revealed relatively rich genetic diversity in the 8 persimmon populations. Analysis of molecular variance (AMOVA) showed that the vari- ance component among populations contributes 24.17% at a significantly high level (P〈0.O01). Further- more, a low level of genetic variation among populations was detected based on the genetic distance (D〈 0.231 7), identity (I〉0.783 2). Neits genetic diversity analysis and AMOVA revealed that there was a higher level of genetic differentiation among populations (~sr= 0.222 6, FST= 0.175 9). [Conclusion]The oriental persimmon populations in Shandong possess abundant genetic diversity, the genetic differentia- tion between populations may result from human selective pressures.
