羟甲香豆素微乳对试验鼠的急性毒性和致突变试验

摘　　要：为探究羟甲香豆素微乳临床应用的安全性,以小白鼠和SD大鼠为试验动物,通过急性毒性试验、微核试验和精子畸形试验观察羟甲香豆素微乳LD50、骨髓细胞微核率和精子畸形率。结果表明,羟甲香豆素微乳的小鼠口服LD50为2.63×104 mg/kg,95%置信区间为2.21×104～3.14×104 mg/kg,毒力回归方程为y=3.57x-0.44(r2=0.977 6),羟甲香豆素微乳的大鼠皮肤给药LD50为6.78×104 mg/kg,95%置信区间为5.94×104～7.73×104 mg/kg,毒力回归方程为y=0.31x-0.43(r2=0.981 3)。根据口服和皮肤用药LD50值判断为无毒级药物,羟甲香豆素微乳对小白鼠骨髓细胞微核率和小白鼠精子的畸形率无明显影响,说明羟甲香豆素微乳没有致突变作用。In order to explore the clinical application security,LD50 of hymecromone microemulsion,bone marrow cell micronucleus rate and sperm malformation rate were observed by acute toxicity test,the micronucleus test and sperm abnormality test in mice.The results showed that the LD50 of Hymecromone microemulsion was 2.63×104 mg/kg by means of oral,its 95% confidence interval was between 2.21×104 mg/kg to 3.14×104 mg/kg,and toxicity regression equation was y=3.57x-0.44（r2= 0.977 6）.The LD50 of Hymecromone microemulsion was 6.78×104 mg/kg by means of skin coating,its 95% confidence interval was between 5.94×104mg/kg to 7.73×104mg/kg and toxicity regression equation was y=0.31x-0.43（r2=0.981 3）.Hymecromone microemulsion is nontoxic drug based on the LD50 of oral or skin coating application test.Hymecromone microemulsion had no significant effects on bone marrow micronucleus rate and sperm abnormality rate,and this means it has no mutagenic effect on mice.
