摘  要:目的通过测定在不同培养时间下大肠杆菌BL21(DE3)中整合子的整合频率,研究饥饿应激对整合子捕获耐药性基因盒频率的影响。方法将整合子和氨基糖苷腺苷转移酶(aadA2)基因盒克隆到同一质粒pACYC184的不同位点,该重组质粒和高表达整合酶的重组质粒同时转化大肠杆菌BL21(DE3),对10株此菌株培养12h和24h后用实时荧光定量聚合酶链反应(PCR)测定发生整合作用的整合子的拷贝数和总的整合子的拷贝数,前者除以后者即为整合频率。结果大肠杆菌BL21(DE3)中整合子在12h整合频率的均值为5.51×10-5,在24h整合频率的均值为1.82×10-4。比较10株此菌株2个时点的整合频率,差异有统计学意义(P<0.01)。结论饥饿应激对整合子捕获耐药性基因盒频率有明显的提高作用。Objective To examine the integron frequency of BL21(DE3) in Escherichia colitis (E. coli) at vari-ous time points during incubation and the effects of hunger stress on integron-capturing antibiotic resistance gene cassettes. Methods The integron and aminoglycoside adenosine transferrase (aadA2) gene cassette were cloned into different sites of the plasmid pACYC184, which was transformed into E.coli BL21 (DE3) strain containing plasmids overexpressing the integrase. This entailed measurement of the copies of integrates and total integron, and the integron frequency as derived from their quotient, following a 12-and 24-hour incubation, respectively. Results Compared with the 12-hour incubation, the 24-hour incubation yielded a remarkably increased integration frequency (1.82×10^-4 vs.5.51×10^-5, P〈0.01) in BL21 (DE3) E. coli. Conclusion Hunger stress may lead to a substantial increase in the frequency of integron-capturing antibiotic resistance gene cassettes.
