摘  要:目的研究GATA-4转染骨髓间充质干细胞(BMSCs)对心肌细胞(CM)保护的影响。方法体外分离培养BMSCs及CM。实验分为常氧培养CM(A)组、低氧培养CM(B)组、低氧+CM+BMSCsNull(C)组和低氧+CM+BMSCsGATA-4(D)组。倒置显微镜下观察CM形态变化,试剂盒检测乳酸脱氢酶(LDH)释放、MTT摄取及膜联蛋白V阳性细胞数。结果与A组相比,B组大部分CM明显收缩变圆,LDH释放增多,MTT摄取减少,膜联蛋白V阳性细胞数增加(P<0.05);而C组和D组能改善B组上述各指标变化过程,其中D组作用更为明显(P<0.05)。结论 GATA-4过表达BMSCs与CM共培养可以通过降低LDH释放、增加MTT摄取和降低膜联蛋白V阳性细胞数来保护CM。Objective To investigate the effects of GATA-4transfection on cardiomyocytes(CM)protected by mesenchymal stem cells(BMSCs)in rats.Methods BMSCs and CMs were isolated and cultured in vitro,and then divided into four groups of A(CM exposed to normoxia),B(CM exposed to hypoxia),C(co-culture of CM and BMSCsNull exposed to hypoxia)and D(co-culture of CM and BMSCsGATA-4 exposed to hypoxia).The morphologic change of CM was observed under inverted microscope.The cell injury markers such as LDH release,MTT uptake and positive cells of annexin-V were detected by kits.Results Compared with group A,the majority of CM were shrank and became round,LDH release and positive cells of annexin-V were increased,while MTT uptake was decreased in group B(P〈0.05),which were all improved in groups of C and D,especially in group D(P〈0.05).Conclusion GATA-4overexpression in co-culture of CM and BMSCs can protect CM via increasing MTT uptake and decreasing LDH release and positive cells of annexin-V.
