摘  要:目的探讨十二指肠空肠旁路术对非肥胖2型糖尿病GK大鼠小肠糖异生相关基因葡萄糖-6-磷酸酶(G-6-P)、磷酸烯醇丙酮酸羧激酶(PEPCK)mRNA表达的影响。方法非肥胖2型糖尿病GK大鼠及正常Wistar大鼠各16只,随机分为GK大鼠手术组(A组)、GK大鼠假手术组(B组)、Wistar大鼠手术组(C组)及Wistar大鼠假手术组(D组),每组各8只。术后第8周取大鼠十二指肠段的小肠组织,RT-PCR检测G-6-P和PEPCK mRNA表达量。结果A组PEPCK和G-6-PmRNA表达量显著高于B组(P<0.01)。C组与D组G-6-P和PEPCK mRNA表达量比较差异无统计学意义。结论十二指肠空肠旁路术能够上调GK大鼠小肠糖异生相关基因G-6-P和PEPCK基因的表达,促进小肠糖异生的发生。Objective To investigate the effect of duodenal-jejunal bypass(DJB)on the mRNA expressions of intestine glucose-6-phosphatase(G-6-P)and phosphoenolpyruvatecarboxylase(PEPCK) in non-obese type 2diabetic rats.Methods A total of 16GK rats and 16 Wistar rats was randomly divided into GK operation(group A),GK sham operation(group B),Wistar operation(group C)and Wistar sham operation(group D)with 8rats each.At the end of 8 weeks after operation,the duodenum of small intestine was taken out for the detections of G-6-P and PEPCK genes mRNA expressions by RT-PCR.Results Compared to group B,PEPCK mRNA expression was significantly increased in group A(P〈0.01).No statistically significant differences of PEPCK and G-6-P mRNA expression were shown between groups of C and D.Conclusion DJB can upregulate G-6-P and PEPCK genes expression of small intestine in GK rats and promote small intestinal gluconeogenesis.
